摘要: This invention relates to an engineered leader-independent heterocyclase (also known as a cyclodehydratase) comprising a defined cyanobactin leader sequence which drives the efficient conversion of heterocyclisable amino acids, such as Ser, Thr and Cys, within a peptide substrate lacking a leader sequence into heterocycles produce a homogenous heterocycle-containing product. This may be useful in biotechnology and chemical synthesis.

摘要: In alternative embodiments, provided herein are transcription/translation (TX-TL) systems and methods of using them for use as rapid prototyping platforms for the synthesis, modification and identification of natural products (NPs), and natural product analogs (NPAs) and secondary metabolites, from biosynthetic gene cluster pipelines. In alternative embodiments, exemplary TX-TL systems as provided herein are used for the combinatorial biosynthesis of natural products (NPs), natural product analogs (NPAs) and secondary metabolites. In alternative embodiments, exemplary TX-TL systems as provided herein are used for the rapid prototyping of complex biosynthetic pathways as a way to rapidly assess combinatorial and biosynthetic designs before moving to cellular hosts. In alternative embodiments, these exemplary TX-TL systems are multiplexed for high-throughput (HT) automation and for prototyping engineered platforms for the synthesis or modification of natural products (NPs), and natural product analogs (NPAs) and secondary metabolites analogs.

摘要: The present application relates to methods of producing exosomes. The application also provides a method for preparing a protein composition comprising culturing an exosome-producing cell expressing a Nef-fusion protein comprising a Nef-derived peptide fused to a protein of interest; isolating exosomes from the exosome-producing cell culture; and purifying the protein of interest from the isolated exosomes. The application further discloses compositions that comprise exosomes containing the Nef-fusion protein, as well as methods of using the Nef-fusion protein and exosomes containing the Nef-fusion protein.

摘要: The present invention relates to a novel beta-galactosidase, and more particularly to a novel beta-galactosidase derived from Bacillus circulans, a gene encoding the beta-galactosidase, a recombinant vector and a recombinant microorganism, which contain the gene, a method for producing a beta-galactosidase using the recombinant microorganism, and a method for producing galactooligosaccharide using the beta-galactosidase. The use of the novel beta-galactosidase according to the present invention makes it possible to efficiently produce a large amount of galactooligosaccharide.

A base sequence 1 for protein expression includes: a gene 3 encoding protein 2; a promoter 4 of the gene 3, the promoter being linked upstream of the gene 3; and a cis element 5 whose activity is improved by an artificial transcription factor 6, the cis element being linked further upstream of the promoter 4. The cis element 5 is represented by SEQ ID NO: 1.

摘要: A platform for preparing a sequence defined biopolymer in vitro is disclosed. The platform includes a ribosome-depleted cellular extract ribosomal RNAs prepared by in vitro transcription and purified ribosomal proteins depleted of ribosomal RNAs. A method of synthesizing and assembling ribosomes in vitro for use in the platform is provided, as well as a method for preparing a sequence defined biopolymer in vitro using assembling ribosomes and the platform.

摘要: The invention relates to a method for producing a lysate used for cell-fee protein biosynthesis, comprising the following steps: a) a genomic sequence in an organism, which codes for an essential translation product that reduces the yield of cell-fee protein biosynthesis, is replaced by the foreign DNA located under a suitable regulatory element, said foreign DNA coding for the essential translation product that additionally contains a marker sequence; b) the organism cloned according to step a) is cultivated; c) the organisms from the culture obtained in step b) are lysed; and d) the essential translation product is eliminated by means of a separation process that is selective for the marker sequence. Also discussed are said lysate and the use thereof.
