摘要: The invention comprises a novel modular, generalizable meso-micro-nano-fluidic platform apparatus, design and methodology which in exemplary embodiments may be applied in conjunction with a novel external triggering and automation/feedback loop control mechanism deployed via computer to explore the phase space of single or double emulsification for applications including the encapsulation of hydrophilic active pharmacological ingredients (APIs). End use applications include the mass production of particulate encapsulation of hydrophobic or hydrophilic APIs with automatic or user-supervised feedback methodology to control and discover mass production or per-drug customized settings of interest for the manufacture of novel or extant therapeutics. This invention allows for a process to produce monodispersed particles of varying sizes and may be used to rapidly screen for optimal size for maximal bioavailability of API particles either on lab bench for in vitro dissolution or in vivo studies, and patient-specific handhelds for maximal drug inhalation.

摘要: The present invention is generally related to systems and methods for producing droplets. The droplets may contain varying species, e.g., for use as a library. In some cases, at least one droplet is used to create a plurality of droplets, using techniques such as flow-focusing techniques. In one set of embodiments, a plurality of droplets, containing varying species, can be divided to form a collection of droplets containing the various species therein. A collection of droplets, according to certain embodiments, may contain various subpopulations of droplets that all contain the same species therein. Such a collection of droplets may be used as a library in some cases, or may be used for other purposes.

摘要: A system, including method and apparatus, for generating droplets suitable for droplet-based assays. The disclosed systems may include either one-piece or multi-piece droplet generation components configured to form sample-containing droplets by merging aqueous, sample-containing fluid with a background emulsion fluid such as oil, to form an emulsion of sample-containing droplets suspended in the background fluid. In some cases, the disclosed systems may include channels or other suitable mechanisms configured to transport the sample-containing droplets to an outlet region, so that subsequent assay steps may be performed.

摘要: A microfabricated sheath flow structure for producing a sheath flow includes a primary sheath flow channel for conveying a sheath fluid, a sample inlet for injecting a sample into the sheath fluid in the primary sheath flow channel, a primary focusing region for focusing the sample within the sheath fluid and a secondary focusing region for providing additional focusing of the sample within the sheath fluid. The secondary focusing region may be formed by a flow channel intersecting the primary sheath flow channel to inject additional sheath fluid into the primary sheath flow channel from a selected direction. A sheath flow system may comprise a plurality of sheath flow structures operating in parallel on a microfluidic chip.
