Abstract: Embodiments disclosed here are directed to novel methods for diagnosing MIS-C or severe acute COVID-19, by detecting a biomarker in a blood sample from a subject suspected of or suffering from MIS-C or severe acute COVID-19, where the methods are for early detection or diagnosis of MIS-C or severe acute COVID-19. Also, disclosed are embodiments directed to apparatuses for use with the novel methods described here. Methods of detecting at least one biomarker and/or diagnosing MIS-C or severe acute COVID-19 disease in a pediatric subject suspected from suffering MIS-C or severe acute COVID-19 that can be used in combination with methods of treating a subject suspected of or suffering from MIS-C or severe acute COVID-19, accelerates the diagnosis and treatment, and improves prognosis.

Abstract: Disclosed is a system and method for a microdevice to separate blood cells based on differences in antigen expression. Specifically, cells of the same phenotype are separated based on whether or not they are activated during infection or resting. The device of the present disclosure takes a small sample of blood and provides differential cell counts that can be used to test for infection and inflammatory response. The device can be used to identify sepsis and other infections rapidly. By measuring differences in activated white cell counts such as neutrophils, the device of the present disclosure measures physiological response to infection in hospitalized patients recovering from burns, surgeries, etc.

Abstract: The present application discloses an electrochemiluminescence immunosensor. The immunosensor includes an electrode functionalized by a nanocomposite film. The film further includes carbon nanohorns dispersed in Nafion® perfluorinated resin solution. The polymeric solution is further stabilized by magnetic nanoparticles. The immunosensor is a Point of care (POC)-based. The immunosensor is configured to work in the range from 100 ng/mL to 1 fg/mL, and has tendency to detect even traces of the tropomyosin. The immunosensor is capable to detect traces even less than 1 fg/mL, hence having high specificity for Tro-Ag detection in food products with distinguished repeatability.

Abstract: This invention relates to an apparatus for conducting immunoassay test. The apparatus includes a groove unit having a groove along a vertical direction configured to hold a rod-shaped portion of a probe along the vertical direction, and a push pin configured to move along a horizontal direction, the push pin being capable of residing at a first position and a second position. A tip of the push pin is capable of pressing the rod-shaped portion of the probe against the groove when the push pin resides at the first position. The distance between the tip of the push pin and the groove is larger than a diameter of the rod-shaped portion of the probe when the push pin resides at the second position.

Abstract: A method of preparing an antibody therapeutic is provided comprising: (a) providing a dissociated cell sample from at least one solid tumor sample obtained from a patient; (b) loading the dissociated cell sample into a microfluidic device having a flow region and at least one isolation region fluidically connected to the flow region; (c) moving at least one B cell from the dissociated cell sample into at least one isolation region in the microfluidic device, thereby obtaining at least one isolated B cell; and (d) using the microfluidic device to identify at least one B cell that produces antibodies capable of binding to cancer cells. The cancer cells can be the patient's own cancer cells. Also provided are methods of treating patients, methods of labeling or detecting cancer, engineered T or NK cells comprising antibodies or fragments thereof, and engineered antibody constructs.
