A porous structure according to the present invention has a polymerase chain reaction (PCT) primer inside pores thereof, and hence, even an inner portion thereof can be used unlike general structures of which only surfaces are used for amplification and detection, thereby maximizing reactivity. In addition, the differentiating of the kinds of primers contained in respective structures leads to detection of several kinds of target nucleic acids at the same and real-time analysis thereof at the same time, and thus is useful for multiplex real-time PCR.

The microfluidic devices include a polymeric medium that includes a first analyte detection domain having a first covalently bound capture member that specifically binds to a first analyte, and a second analyte detection domain having a second covalently bound capture member that specifically binds to a second analyte. Also provided are methods of using the subject microfluidic device, systems and kits that use the subject microfluidic devices, as well as methods of producing the same.

The embodiments disclosed herein are directed to an apparatus useful in conducting detection of compounds on blotting membranes. The device is comprised of several layers including a porous support layer below the blotting membrane(s), a flow distributor above the blotting membrane(s) and optionally a well on the flow distributor to contain the liquid to the desired area and to allow for lower starting volumes of such liquid. Preferably, the flow distributor is a non-binding or low binding hydrophilic porous membrane such as a 0.22 micron membrane and the support layer is a grid or sintered porous material. The distributor and support are held together to form an envelope around the membrane(s). The use of a hinge, clips and other such devices is preferred in doing so.

The present disclosure provides fully integrated microfluidic systems to perform nucleic acid analysis. These processes include sample collection, nucleic acid extraction and purification, amplification, sequencing, and separation and detection. The present disclosure also provides optical detection systems and methods for separation and detection of biological molecules. In particular, the various aspects of the invention enable the simultaneous separation and detection of a plurality of biological molecules, typically fluorescent dye-labeled nucleic acids, within one or a plurality of microfluidic chambers or channels. The nucleic acids can be labeled with at least 6 dyes, each having a unique peak emission wavelength. The present systems and methods are particularly useful for DNA fragment sizing applications such as human identification by genetic fingerprinting and DNA sequencing applications such as clinical diagnostics.

An apparatus for distributing a hydrophobic material in a hydrophilic substrate includes a first plate, a second plate, a heater operatively connected to the first plate and configured to heat the first plate to a first temperature that is greater than a second temperature of the second plate, and an actuator configured to move the first plate and second plate together with a hydrophilic substrate between the plates. The first plate and second plate form a temperature gradient to enable a hydrophobic material on one side of a hydrophilic substrate that faces the second plate to penetrate into the hydrophilic substrate towards the first plate.
