摘要: Described is an agent for inducing viral vector production which includes a cell growth inhibitor A. The cell growth inhibitor A includes a compound which inhibits progression of the cell cycle in G2 phase or M phase. Additionally, a method of producing a viral vector and a method of inducing viral vector production using the same are described. The methods include introducing a nucleic acid into a cell and adding the cell growth inhibitor A to the cell between 6 hours before and 6 hours after the time of introducing the nucleic acid.

摘要: The present invention relates to a cell culture medium for preparing liver, gastric, pancreatic, colon or intestinal adult stem cell isolated from adult tissue, as well as for maintaining such stem cell in the undifferentiated state. The cell culture medium comprises a base medium; an ABL and SRC dual kinase inhibitor/an ABL kinase inhibitor and a SRC kinase inhibitor; a mitogenic factor; a WNT signalling pathway activator; a stimulator for NAD+ and NADP+ generation; and a cAMP/PKA pathway activator. In a particular embodiment, the ABL and SRC dual kinase inhibitor is Dasatinib; the mitogenic factor is EGF; the WNT signalling pathway activator is R-Spondin 1; the stimulator for NAD+ and NADP+ generation is nicotinamide; and the cAMP/PKA pathway activator is cholera endotoxin.

摘要: Described herein are cells, cell culture methods, and cell culture media compositions useful for producing and maintaining iPSC-derided cell lines that are of higher purity and maintain cell type integrity better than current iPSC-derived cell lines. Also disclosed are methods of using the described cells and media, such as therapeutic methods of use for the described cells. The described cells include iPSC-derived mesodermal precursor cells (MPC), which itself may differentiate into at least four different cell types. When cultured under appropriate conditions, the mesodermal precursor cells can be used to produce hematopoietic stem cells (HSC), mesenchymal stem cells (MSC), smooth muscle cells (SMC), or unlimited functional endothelial cells (UFEC). One characteristic that makes the described cells desirable is that they can be maintained in culture for a number of days, or passages, without changing phenotype through differentiation.

摘要: The present invention relates to a method for producing a spheroid including neurons and astrocytes, the method including: culturing induced pluripotent stem cell-neural progenitor cells (iPSC-NPCs) in an NPC medium; culturing the cultured cells in a neuronal differentiation medium (NDM); and culturing the cultured cells in a glial differentiation medium (GDM). The spheroid produced by the method according to the present invention contains mature neurons and astrocytes which express a desired marker after 3 weeks of in vitro culture and has a uniform size, and thus can be used for simple visual screening to confirm drug efficacy.
