摘要: A production method for highly proliferating cells or ectodermal progenitor cells, the production method including (i) preparing ectodermal cells which serve as a raw material, (ii) culturing the ectodermal cells which serve as the raw material in a medium containing a small-molecule signaling pathway inhibitor by bringing the inhibitor into contact with the ectodermal cells, and (iii) after the contact, performing additional culturing in a medium containing the inhibitor to obtain a culture containing highly proliferating cells that have higher cell proliferative ability than the ectodermal cells which serve as the raw material; highly proliferating cells having characteristics of ectodermal cells and proliferative ability that allows the number of the highly proliferating cells after more than 28 days of a culturing period during which the cells are brought into contact with a small-molecule signaling pathway inhibitor to be greater than 1.0 times the number of ectodermal cells.

摘要: The present invention relates to a method for producing a spheroid including neurons and astrocytes, the method including: culturing induced pluripotent stem cell-neural progenitor cells (iPSC-NPCs) in an NPC medium; culturing the cultured cells in a neuronal differentiation medium (NDM); and culturing the cultured cells in a glial differentiation medium (GDM). The spheroid produced by the method according to the present invention contains mature neurons and astrocytes which express a desired marker after 3 weeks of in vitro culture and has a uniform size, and thus can be used for simple visual screening to confirm drug efficacy.

摘要: The invention relates to a cell sheet construct for neurovascular reconstruction. The cell sheet construct has a vascular endothelial cell layer and a neural stem cell layer, and the two layers are physically in direct contact with each other, where the vascular endothelial cell layer forms branching vasculatures, and the neural stem cell layer differentiates into neurons. The invention also relates to a method for manufacturing the cell sheet construct, having the following steps: culturing vascular endothelial cells on a substrate to form a vascular endothelial cell layer, seeding neural stem cells on the vascular endothelial cell layer to make the neural stem cells be physically in direct contact with the vascular endothelial cell layer, and culturing the neural stem cells and the vascular endothelial cell layer to differentiate into neurons and branching vasculatures to form a cell sheet construct.

摘要: The present disclosure relates to the use of laminin-521 in obtaining retinal pigment epithelium (RPE) cells. Pluripotent human embryonic stem cells are cultured on plates coated with recombinant laminin-521 (laminin-11), in totally defined and xeno-free conditions. A first cell culture medium contains a growth factor, and a second cell culture medium does not contain growth factor. The stem cells are first exposed to the first cell culture medium, then exposed to the second cell culture medium for a longer time period. After a number of weeks, clinical grade RPE cells are obtained from the stem cells.
