摘要: Disclosed herein are genetically modified plants having altered biological activity of 3-β-hydroxysteroid dehydrogenase/isomerase (GAME25), or 2-oxoglutarate-dependent dioxygenase (GAME31), or a combination thereof, wherein the genetically modified plants have an altered content of at least one cholesterol derived compound selected from the group including a steroidal alkaloid or a glycosylated derivative thereof and an unsaturated or saturated steroidal saponin or a glycoside derivative thereof. Further disclosed herein are genetically modified plants having altered expression of a gene encoding a 3-β-hydroxysteroid dehydrogenase/isomerase (GAME25), or a 2-oxoglutarate-dependent dioxygenase (GAME31), or a combination thereof, wherein the genetically modified plant has an altered content of at least one cholesterol derived compound selected from the group including a steroidal alkaloid or a glycosylated derivative thereof and an unsaturated or saturated steroidal saponin or a glycoside derivative thereof. Methods of producing these genetically modified plants are also disclosed.

摘要: The present invention relates to the field of microbial production of novel biosurfactants. More specifically, the present invention discloses the usage of a fungal strain such as the yeast Starmerella bombicola having a dysfunctional CYP52M1 cytochrome P450 monooxygenase and a dysfunctional FAO1 fatty alcohol oxidase for producing high amounts of so-called “symmetrical bolaform sophorosides” where both sophorose moieties are attached through a terminal glycosidic linkage to the hydrophobic linker. In addition, the present invention further discloses that the latter yeast can also be used to produce alkyl sophorosides and symmetrical bolaform glucosides.

摘要: The present disclosure provides a recombinant Escherichia coli for producing chlorogenic acid and application thereof. In the present disclosure, tyrosine ammonia-lyase FjTAL derived from Flavobacterium johnsoniae, hpaBC derived from E. coli, 3-deoxy-D-arabino-heptulosonate-7-phosphate synthase mutant aroGfbr, chorismate mutase tyrC derived from Zymomonas mobilis, quinic acid/shikimate-5 dehydrogenase ydiB derived from E. coli, hydroxycinnamoyl CoA:quinic acid transferase NtHQT derived from Nicotiana tabacum, and 4-coumarate:CoA ligase At4CL1 derived from Arabidopsis thaliana are expressed in the recombinant E. coli, thereby constructing a chlorogenic acid biosynthesis pathway in E. coli. Then, the aroB gene and gldA gene derived from E. coli are overexpressed, and an endogenous gene menI is knocked out from the recombinant E. coli. The recombinant strain produced chlorogenic acid by fermentation at a titer of up to 638.2 mg/L in a shake flask or at a titer of 2.8 g/L in a 5-L fermenter.

摘要: A cofactor self-sufficient Escherichia coli and its construction method and application in the synthesis of L-glufosinate are provided. The present invention expresses a NADH kinase and key enzymes of the cofactor synthesis pathway in E. coli, and knocks out the genes of enzymes that catabolizes cofactor, and with the addition of co-metabolic intermediates during cell incubation, the intracellular NADP(H) concentration is increased by at least 50% and the catalytic activity of glufosinate dehydrogenase by 2-fold, resulting in a significant increase in the spatiotemporal yield of the glufosinate synthesis reaction.

摘要: The present invention relates to the technical field of microorganisms, and specifically to engineered microorganisms expressing acetoacetyl-CoA reductase variants and methods for increasing the proportion of 3-hydroxyhexanoic acid in PHA. The acetoacetyl-CoA reductase variants and their coding genes provided by the present invention can significantly increase the proportion of 3-hydroxyhexanoic acid in PHA produced by strains; the proportion of 3-hydroxyhexanoic acid in PHA produced by the engineered Ralstonia eutropha constructed utilizing the acetoacetyl-CoA reductase variants and their coding genes provided by the present invention is significantly increased, which provides new genes and strain resources for strains producing poly(3-hydroxybutyrate-co-3-10 the development of engineered hydroxyhexanoate) with high proportion of 3-hydroxyhexanoic acid.
