摘要: In related-art methods of differentiating pluripotent stem cells into a desired cell type, there has not been established a differentiation induction method using human ES/iPS cells and being highly efficient. Many attempts have been made, including a stepwise differentiation induction method based on the control of culture conditions or the addition of, for example, various cell growth factors/differentiation factors to a culture solution, but the use of complicated culture steps is a big problem. A method of inducing differentiation into a desired cell type within a short period of time and with high efficiency by use of a pluripotent stem cell that actively undergoes cell differentiation, which is obtained by reducing an undifferentiated state of the pluripotent stem cell, has been developed, and thus the present invention has been completed.

摘要: The present invention provides a method for producing a composition for culturing animal cells, wherein the method includes: (1) a step in which an algae is mixed with a solid acid catalyst and an algae extract is obtained by heating; and (2) a step in which the algae extract is added to a medium for culturing animal cells and the concentration of the algae extract is adjusted. The present invention also provides a recycling/culturing method for algae and animal cells including: (i) a step in which a waste liquid (a first waste liquid) previously used to culture animal cells is used to culture algae; (ii) a step in which the algae is collected, mixed with a solid catalyst, and heated, and an algae extract is thereby obtained; (iii) a step in which the algae extract is added to the waste liquid (a second waste liquid) previously used to culture algae in (i), the concentration of the algae extract is adjusted, and a composition for culturing animal cells is produced; and (iv) a step in which animal cells are cultured using the composition for culturing animal cells.

摘要: Forced expression of a handful of transcription factors (TFs) can induce conversions between cell identities; however, the extent to which TFs can alter cell identity has not been systematically assessed. Here, we assembled a human TFome, a comprehensive expression library of 1,578 human TF clones with full coverage of the major TF families. By systematically screening the human TFome, we identified 77 individual TFs that induce loss of human-induced-pluripotent-stem-cell (hiPSC) identity, suggesting a pervasive ability for TFs to alter cell identity. Using large-scale computational cell type classification trained on thousands of tissue expression profiles, we identified cell types generated by these TFs with high efficiency and speed, without additional selections or mechanical perturbations. TF expression in adult human tissues only correlated with some of the cell lineage generated, suggesting more complexity than observation studies can explain.
