摘要: A system and method for automatically digesting, culturing and growing a biopsy. The digester is a closed fluid transfer system, including disposable and durable parts, that receives a biopsy, washes the biopsy tissue, digests the tissue to separate the cells from each other, and seeds the cells in a culture flask. In one use, the system receives cells from a patient, expands and multiplies them, and returns them to the patient. A controller coordinates the motion of a plurality of pumps and the status of a plurality of valves, the pumps and the plurality of valves moving the fluid throughout the system. A sensor system assists the controller in volume and flow rate control of the fluids in the system, and in controlling the position of a tube providing air and fluids to a biopsy vial that includes the biopsy.

The present disclosure can realize the whole anaerobic operation of urine collection and culture in the bladder.

摘要: A portable apparatus useful for collection and processing of human biological material, such as adipose or cancellous bone material, to prepare a concentrated product (e.g., stromal vascular fraction). The apparatus has a container with a containment volume with a tissue retention volume and a filtrate volume separated by a filter and with a pellet well for collecting concentrate product in the form of a pellet phase from centrifuge processing. The pellet well is accessible only from above when the apparatus is in an access orientation. Collected pellet phase material may be removed from the pellet well by direct aspiration, without suspending the material in a suspension liquid within the container. Access ports may be configured for access only from above the container. The apparatus may include a tissue collector disposed in the disuse retention volume to engage and collect collagen or other stringy tissue. A method of processing adipose tissue to concentrate leuko stromal vascular cells includes multi-step processing using a portable container.

The plunger may be configured to be actuated to open the seal and express the portion of the sample.

摘要: The present invention provides automated devices for use in supporting various cell therapies and tissue engineering methods. The present invention provides an automated cell separation apparatus capable of separating cells from a tissue sample for use in cell therapies and/or tissue engineering. The cell separation apparatus can be used in combination with complementary devices such as cell collection device and/or a sodding apparatus to support various therapies. The automated apparatus includes media and tissue dissociating chemical reservoirs, filters, a cell separator and a perfusion flow loop through a graft chamber which supports a graft substrate or other endovascular device. The present invention further provides methods for using the tissue grafts and cell samples prepared by the devices described herein in a multitude of therapies including revascularization, regeneration and reconstruction of tissues and organs, as well as treatment and prevention of diseases.

摘要: The present invention efficiently and cost-effectively extracts and collects cells from a tissue. The inventors have discovered that the tissue can be effectively fragmented and the resulting cells can be purified using a system or kit with multiple components. An advantage of the present invention is that tissue processing takes place in a closed system such that sterility can be maintained throughout the process, even if certain components are removed during processing, for example through the use of valves, clamps, and heat seals. Furthermore, any or all of the steps can be automated or manually accomplished, according to the specific needs of the application or the user.

摘要: A tissue dissociation method and system for washing and dissociating tissue and producing a cellular suspension for use in autologous, cell-based therapies or research wherein a sterile environment is maintained. More particularly, a system for dissociating tissue comprising a housing unit, a first compartment having a plurality of pores providing for fluid communication between the interior of the first compartment and the interior of the housing unit, a first inlet port that provides communication between the exterior environment and the interior of the first compartment and thereby provides a means for introducing solutions into the interior of the first compartment, a second compartment, a second inlet port for introducing a tissue sample into the system, a means for agitating or rotating the second compartment in the housing unit, a means for controlling the exit of effluent from the housing unit, a first receptacle, and a centrifuge-ready second receptacle for collecting a resulting cell suspension. The method comprises introducing a tissue sample into the system and subjecting it to a washing phase and a dissociating phase wherein the tissue is agitated or rotated. The tissue may also be subjected to a sedentary phase and a phase in which a solution is added to the resulting cell suspension to cease tissue dissociation. At the conclusion of the tissue dissociation process, the resulting cell suspension is collected in a centrifuge-ready receptacle.
