This disclosure provides systems, methods, and devices for processing samples on a microfluidic device. One method includes moving a sample from an upstream channel of a microfluidic device into a DNA manipulation module located downstream of the upstream channel. The DNA manipulation module includes a DNA manipulation zone configured to perform amplification of the sample, a first valve disposed upstream of the DNA manipulation zone, and a second valve disposed downstream of the DNA manipulation zone. The method also includes receiving the sample in the DNA manipulation zone; closing the first valve and the second valve such that as and liquid are prevented from flowing into or out of the DNA manipulation zone; and thermal cycling the sample in the DNA manipulation zone.

An analyzing cartridge and method for using it to analyze a sample having a plurality of reservoirs and capillaries connected for communication between these reservoirs and is provided therein with a reagent required for analysis. The reservoirs are provided with openings leading to the outside of the cartridge, and the openings are covered with vents each consisting of a gas-permeable/non-liquid-permeable, hydrophobic, porous membrane. The analyzing cartridge requires only trace amounts of a sample and a reagent and no maintenance. It is provided with a non-fluid reagent in vent-carrying reservoirs thereby making it possible to formulate a very trace amount of reagent solution in the cartridge by injecting a reagent dissolving liquid into the reservoirs immediately before analyzing. A liquid feed control device controls the feeding of the liquid between the reservoirs via the capillaries when it is attached to the cartridge to allow or control the entry/exit of air via the vents.

The present invention relates to a system and method for moving samples, such as fluid, within a microfluidic system using a plurality of gas actuators for applying pressure at different locations within the microfluidic. The system includes a substrate which forms a fluid network through which fluid flows, and a plurality of gas actuators integral with the substrate. One such gas actuator is coupled to the network at a first location for providing gas pressure to move a microfluidic sample within the network. Another gas actuator is coupled to the network at a second location for providing gas pressure to further move at least a portion of the microfluidic sample within the network. A valve is coupled to the microfluidic network so that, when the valve is closed, it substantially isolates the second gas actuator from the first gas actuator.

Improved microfluidic devices, systems, and methods allow selective transportation of fluids within microfluidic channels of a microfluidic network by applying, controlling, and varying pressures at a plurality of reservoirs. Modeling the microfluidic network as a series of nodes connected together by channel segments and determining the flow resistance characteristics of the channel segments may allow calculation of fluid flows through the channel segments resulting from a given pressure configuration at the reservoirs. To effect a desired flow within a particular channel or series of channels, reservoir pressures may be identified using the network model. Viscometers or other flow sensors may measure flow characteristics within the channels, and the measured flow characteristics can be used to calculate pressures to generate a desired flow. Multi-reservoir pressure modulator and pressure controller systems can optionally be used in conjunction with electrokinetic or other fluid transport mechanisms.

A device for dispensing liquids includes one or more liquid outlet means carried on a movable head. The device forms part of an automated excision apparatus for cutting biomolecule spots from an array of such spots carried in a gel. The head is mounted on an automated motion control system to allow for movement of the head in X, Y, Z directions under the control of a control means. A displacement means for causing the dispensing of liquid from the one or more liquid outlet means of the device is also carried by the moveable head carrying the liquid delivery outlets allowing the length of the tubing between the outlet and the aspiration device, typically a syringe, to be much shorter. This means that swelling and contraction of the tube is minimized during liquid handling operations which means more accurate delivery of specified volumes through the outlet. The syringes and the outlets are mounted on opposite sides of an axis of movement of the apparatus, keeping the machine compact. Typically the axis will be a horizontal X or vertical Y axis defined by a beam or the like which supports the movable head.

A multi-channel fluid dispenser includes a reservoir with multiple internal chambers, a multi-channel liquid dispensing head, a plurality of fluid-delivery conduits, and support and positioning elements. The fluid-delivery conduits receive liquid from the chambers in the reservoir and deliver it to the dispensing head. The support and positioning elements are operable to support and position the dispensing head so that it can deliver the liquid from the reservoir to an underlying receiver. The fluid-delivery conduits include one or more optional flow control features that improve the accuracy of the dispensing operation. The multi-channel liquid dispensing head, which includes a valve-support member, a plurality of valves and a plurality of nozzles, is advantageously movable in three directions to facilitate alignment with an underlying receiver. Movement of the dispensing head along two axes is provided by the simple expedient of elongate holes having their long axes aligned in mutually orthogonal directions. In some embodiments, the valves are tiltable away from the vertical so that liquid dispensed from such tilted valves is directed at a non-normal angle into a receiver.
