摘要: A system and method for preparing a cancer vaccine (and optionally purifying the blood) has a blood filtration system, controlled by a processing unit, for filtering exogenous blood plasma to isolate tumor cells, tumor stem cells and tumor breakdown products. The blood filtration system filter may include multiple layers having differently sized apertures to retain differently sized materials (from among tumor cells of different sizes, tumor stem cells and tumor breakdown products. A device directs electromagnetic radiation at the separated tumor cells, tumor stem cells and/or tumor breakdown products. The electromagnetic radiation may cause the separated tumor cells, separated tumor stem cells and/or separated tumor breakdown products (for example tumor protein such as DNA and/or tumor exosomes) to have a coagulated outer layer such as by degrading the outer surface. The electromagnetic radiation may have a UV wavelength. A conical coil improves flow rate uniformity. Tumor exosomes may be centrifuged.

摘要: This disclosure relates to adsorbent compositions and methods for selective capture, purification, and release of marker cells for cancer, more particularly, circulating rare cells exemplified by circulating tumour cells followed by their isolation as single cells. These adsorbent compositions include functionalized substrates linked to the ligands which bind non-covalently to a marker cell for cancer, through at least one linkage cleaved at controlled rate in the presence of specific stimuli, and also linked to ligands which bind non-covalently to leukocytes through non-cleavable linkages. The capture of marker cell for cancer and release may be monitored using microscopy. Methods for the capture and controlled release of these marker cells using these adsorbent compositions lead to isolation of single cells useful in diagnosis, prognosis and in the screening of therapeutic treatment of diseases, especially oncological diseases, genomics, single cell whole genome amplification, multi-omics, transcriptomics, proteomics, to identify genetic, genomic signatures, mutations.

摘要: The disclosure relates to an apparatus for segregating particles on the basis of their ability to flow through a stepped passageway. At least some of the particles are unable to pass through a narrower passageway bounded by a segregating step, resulting in segregation of the particles. The breadth of the leading edge of at least one step of the apparatus is significantly greater than the overall width of the passageway in which the step occurs, permitting high and rapid sample throughput. The apparatus and methods described herein can be used to segregate particles of a wide variety of types. By way of example, they can be used to segregate circulating tumor cells from a human blood sample.
