Abstract:
The present invention relates to methods of treating and preventing Staphylococcus aureus infection and/or a condition resulting from a S. aureus infection a subject that involves administering a CD11b inhibitor. The present invention further relates to a non-human transgenic animal expressing human CD11b and its use in methods of identifying novel therapeutics for the treatment and prevention of Staphylococcus aureus infection and/or a condition resulting from a S. aureus infection.
Abstract:
An effective Staphylococcus aureus vaccine may require several antigenic components, and so various combinations of S. aureus antigens are identified for use in immunisation. These polypeptides may optionally be used in combination with S. aureus saccharides.
Abstract:
Disclosed are methods and compositions for treating or preventing a Staphylococcus bacterial infection using a non-toxigenic Protein A (SpA) variant.
Abstract:
Disclosed are novel immunogenic proteins derived from Staphylococcus aureus, as well as methods for their use in conferring protective immunity against S. aureus infections. Also disclosed are nucleic acids encoding the proteins and methods of use of these nucleic acids.
Abstract:
The present invention relates to peptides of the enolase protein from Staphylococcus aureus as well as nucleic acid and nucleic acid sequence homologs encoding the peptides. The present invention also relates to a composition, particularly a S. aureus vaccine, comprising one or more of the enolase peptides described herein or a fragment, derivative or variant thereof capable of generating an immune response that induces a protective antibody response or opsonophagocytic activity of human neutrophils for S. aureus. The present invention also encompasses methods of treating and/or reducing the likelihood of a Staphylococcus infection by administering a composition of the invention.
Abstract:
The present invention relates to methods of inducing an immune response to Staphylococcus comprising administering a composition comprising an SA2493-related polypeptide from Staphylococcus aureus as well as derivatives or fragments thereof. The present invention also encompasses methods of treating and/or reducing the likelihood of a Staphylococcus infection by administering a composition comprising an SA2493-related polypeptide or an antibody that specifically binds to an SA2493 polypeptide, derivative or fragments thereof. Compositions administered in the methods of the invention can include one or more additional antigens including, but not limited to, IsdB. Compositions used to practice the methods of the invention are also encompassed.
Abstract:
The present invention relates to chromatography matrices including ligands based on one or more domains of immunoglobulin-binding proteins such as, Staphylococcus aureus Protein A (SpA), as well as methods of using the same.
Abstract:
The present invention concerns methods and compositions for treating or preventing a bacterial infection, particularly infection by a Staphylococcus bacterium. The invention provides methods and compositions for stimulating an immune response against the bacteria. In certain embodiments, the methods and compositions involve a non-toxigenic Protein A (SpA) variant.
Abstract:
A protein structure capable of selective interaction with an organic target is provided. The protein structure is a polymer comprising as a repeating structural unit a recombinant fusion protein that is capable of selective interaction with the organic target. The fusion protein is comprising the moieties B, REP and CT, and optionally NT. B is a non-spidroin moiety of more than 30 amino acid residues, which provides the capacity of selective interaction with the organic target. REP is a moiety of from 70 to 300 amino acid residues and is derived from the repetitive fragment of a spider silk protein. CT is a moiety of from 70 to 120 amino acid residues and is derived from the C-terminal fragment of a spider silk protein. NT is an optional moiety of from 100 to 160 amino acid residues and is derived from the N-terminal fragment of a spider silk protein. The fusion protein and protein structure thereof is useful as an affinity medium and a cell scaffold material.
Abstract:
The present invention relates to a chromatography ligand, which comprises Domain C from Staphylococcus protein A (SpA), or a functional fragment or variant thereof. The chromatography ligand presents an advantageous capability of withstanding harsh cleaning in place (CIP) conditions, and is capable of binding Fab fragments of antibodies. The ligand may be provided with a terminal coupling group, such as arginine or cysteine, to facilitate its coupling to an insoluble carrier such as beads or a membrane. The invention also relates to a process of using the ligand in isolation of antibodies, and to a purification protocol which may include washing steps and/or regeneration with alkali.
