摘要: Methods, assays, and compositions for identifying molecular subtypes of metastatic cancer are disclosed. Methods include determining expression levels of genes and/or miRNAs in a sample of metastatic tissue and identifying the molecular subtype of the metastasis based on the determined expression levels using a neural network-based classifier. Methods may further include providing a prognosis and making treatment decision based on the molecular subtype of the metastasis.

摘要: Novel means that enables detection of the monocytogenes bacterium alone distinctly from other bacteria belonging to the genus Listeria with sufficiently high accuracy is disclosed. The present inventors intensively analyzed the genome of the monocytogenes bacterium to identify two genes (the lmo0084 gene and the lmo2736 gene) as target regions with which the monocytogenes bacterium can be specifically detected distinctly from other bacteria belonging to the genus Listeria utilizing a nucleic acid amplification method. By a further intensive study of the base sequences of these two genes, primer setting regions for highly accurate, specific detection of the monocytogenes bacterium alone were identified, and preferred particular examples of PCR primer sets, LAMP primer sets, and real-time PCR primer-probe sets were established.

摘要: The disclosure pertains to a method for isolating DNA fragments that contain a binding site for a guide-RNA of an RNA-guided endonuclease. The method comprises contacting the RNA-guided endonuclease with a target double-stranded DNA, thereby producing a single stranded DNA (ss-DNA) at the site to which the guide-RNA binds. The ssDNA is then reacted with kethoxal or an analog thereof modified by a chemoselective group, thereby adding the chemoselective group to any unpaired guanine based within the ssDNA. A first binding member of a specific binding pair is linked to the chemoselective group, for example, via a cycloaddition reaction. The DNA is fragmented and the DNA fragments that contain the binding sites for the guide-RNA are enriched using a support that comprises a second binding member of the specific binding pair. The method can be used to identify binding sites for a guide-RNA thereby identifying off-target binding sites.

摘要: A microfluidic analysis system (1) performs polymerase chain reaction (PCR) analysis on a bio sample. In a centrifuge (6) the sample is separated into DNA and RNA constituents. The vortex is created by opposing flow of a silicon oil primary carrier fluid effecting circulation by viscous drag. The bio sample exits the centrifuge enveloped in the primary carrier fluid. This is pumped by a flow controller (7) to a thermal stage (9). The thermal stage (9) has a number of microfluidic devices (70) each having thermal zones (71, 72, 73) in which the bio sample is heated or cooled by heat conduction to/from a thermal carrier fluid and the primary carrier fluid. Thus, the carrier fluids envelope the sample, control its flowrate, and control its temperature without need for moving parts at the micro scale.
