Also provided herein are methods of making and using such cells, for example to measure the ability of a chimeric antigen receptor (CAR) to induce nuclear factor of activated T cells (NFAT)-signaling in a cell.

摘要: This invention pertains to isolated mutant Cas9 nucleic acids and proteins for use in CRISPR/Cas endonuclease systems and their methods of use. In particular, the invention pertains to an isolated mutant Cas9 protein, wherein the isolated mutant Cas9 protein is active in a CRISPR/Cas endonuclease system, wherein the CRISPR/Cas endonuclease system displays a lower ratio of reduced off-target editing activity to on-target editing activity for at least one target site relative to the corresponding ratio of reduced off-site editing activity to on-target editing activity of a wild-type CRISPR/Cas endonuclease system having a WT-Cas9 protein of SEQ ID NO:5.

摘要: The present disclosure provides methods to identify peptides and small molecule moieties that are able to modulate protein-protein interactions (PPIs). Some moieties can disrupt specific PPIs within a complex, or disrupt variant-specific PPIs. Some moieties can alternatively bridge between two proteins in a protein-specific or a variant-specific manner. The methods described enable generation of compounds able to modulate PPI networks within cells with implications for drug development for pathological conditions.

A genetically modified bacteriophage is also disclosed which comprises an indicator gene in the late gene region. The specificity of the bacteriophage, such as Salmonella-specific bacteriophage, allows detection of a specific microorganism, such as Salmonella spp. and an indicator signal may be amplified to optimize assay sensitivity.

摘要: An NADP(H) nanosensor has i) a nucleic acid sequence to which a regulator is capable of binding, wherein the oxidation state of the regulator depends on the NADP(H) availability; ii) a promoter sequence following the nucleic acid sequence i), to which an RNA polymerase is capable of binding, wherein the affinity of the RNA polymerase for the promoter sequence is influenced by the oxidation state of the regulator; iii) a nucleic acid sequence which is under the control of the promoter sequence ii) and which codes for an autofluorescent protein. The present invention also relates to a cell, a method for isolating genes which code for NADP(H)-dependent enzymes, and the use of an NADP(H) nanosensor.
