摘要: Disclosed herein are methods and systems for rapid detection of microorganisms such as Cronobacter spp. in a sample. A genetically modified bacteriophage is also disclosed which comprises an indicator gene in the late gene region. The specificity of the bacteriophage, such as Cronobacter-specific bacteriophage, allows detection of a specific microorganism, such as Cronobacter spp. and an indicator signal may be amplified to optimize assay sensitivity.

These gene constructs and expression vectors are useful for making a mammalian cell incorporating the Cypridina luciferase gene and the copepod luciferase to be capable of stably expressed and extracellularly secreted under the control of the distinct promoters.

摘要: Assays for detecting potentially toxicdanalytes utilize a preparation of eukaryotic or prokaryotic cells which have signal-generating metabolic activity. The sensitivity of such assays is significantly improved by the employment of a particulate solid-phase adsorbent e.g. silica gel, which can be preliminarily mixed either with the suspected toxic analyte or with the cellular reagent. It is found that the presence of either the adsorbent-analyte combination or the adsorbent-cellular reagent combination results in an amplification of the decrease of the intensity of the generated signal, thereby improving the detection capability of the assay, particularly with respect to low concentrations of toxicants.

摘要: A method for detection, identification and/or quantification of bacteriophage of bacterial host specificity for bacterial genus, species or serotype, based upon the occurrence of release of cell contents, particularly nucleotides e.g. ATP, on lysis of bacterial cell walls on incubation with bacterial host cells. When new phage particles are released at the end of the phage replication cycle nucleotide levels are measured and compared with controls. The method provides for the detection of specific phages which is faster and more sensitive than known techniques. The method is only limited by the availability of host bacteria/target phage pairings.

摘要: A method for detecting the presence of a lysed eukaryotic cell in a sample, said method comprising: (i) adding adenosine diphosphate (ADP) to said sample under conditions which allows the conversion of ADP to adenosine triphosphate (ATP) by cellular adenylate kinase, (ii) detecting ATP in said sample and relating that to the presence of adenylate kinase and thus to the presence of lysed cells. The method is useful in detecting the cell lysis, for example when screening for drugs which are required to cause lysis, for example for use in tumor therapy. However, in addition, cells may, in a preliminary step, be lysed and the contents quantitated in order to establish for example the health of condition of the cells or to detect the presence of cells in sample such as milk or urine, for diagnostic purposes.
