Also provided is a method of replacing a tooth in the mouth of a mammal, where the tooth is absent and a tooth socket is present in the mouth at the position of the absent tooth. The method includes implanting an acellular scaffold having the shape of the missing tooth into the tooth socket. Additionally, a method of making a tooth scaffold is provided. The method includes synthesizing an acellular scaffold in the shape of a mammalian tooth and adding at least one compound that is chemotactic, osteogenic, dentinogenic, amelogenic, or cementogenic.

Calcium-phosphate nanofiber matrices comprising randomly dispersed crystalline calcium-phosphate nanofibers are provided. The nanofibers are synthesized using sol-gel methods combined with electrospinning. The nanofibers may be hollow, solid or may comprise a calcium-phosphate shell surrounding a polymer containing inner core to which biologically functional additives may be added. The nanofiber matrices may be used to culture bone and dental cells, and as implants to treat bone, dental or periodontal diseases and defects.

Porous HCG scaffolds are provided in a perfusion bioreactor. Cells are seeded in the HCG scaffolds and cell culture media is perfused through the bioreactor to allow for cell seeding and growth. The cell culture media is removed, the HCG-cell constructs washed, and then preserved in the bioreactor with a perfusion comprising cryopreservation fluid comprising one or more of DMSO, trehalose, glycerol, ethylene glycol, or serum. The HCG-cell constructs (or the perfusion chambers containing them) are then removed from the bioreactor and placed in a cryopreservant media and maintained at about −80° C. The frozen HCG-cell constructs (or the chambers containing them) can then be stored at a suitable cryogenic temperature until needed. When needed, frozen HCG-cell constructs can be removed from cold storage and thawed using suitable means.

An implant for articular cartilage repair includes (1) a three-dimensional body formed of cancellous bone having a demineralized section that contains bone morphogenetic proteins (BMP's) that are released by the demineralization but retained in the body, and (2) a cartilage layer formed on a surface of the demineralized section. The cartilage layer is formed by a method including the steps of (a) isolating chondrocytes from articular cartilage of a donor; (b) cultivating the isolated chondrocytes in a medium; (c) adding the cultivated chondrocytes to the demineralized section of the body, whereby the cultivated chondrocytes are stimulated by the BMP's retained in the body; and (d) incubating the cultivated chondrocytes to form a plurality of layers of chondrocytes on the demineralized section, wherein the plurality of layers of chondrocytes forms the cartilage layer.
