摘要: A method for in-vitro production of a cohesive cartilage construct includes the following steps: a) propagating chondrogenic cells derived from a subject to allow formation of one or more cartilage micro constructs; b) putting a plurality of the cartilage micro constructs in motion to facilitate contact between the cartilage micro constructs and thereby allow formation of one or more fused cartilage micro constructs; and c) subjecting one or more of the fused cartilage micro constructs to mechanical stimulation in a hypoxic environment to allow formation of a cohesive cartilage construct.

摘要: Methods and compositions for the biological repair of cartilage using a hybrid construct combining both an inert structure and living core are described. The inert structure is intended to act not only as a delivery system to feed and grow a living core component, but also as an inducer of cell differentiation. The inert structure comprises concentric internal and external and inflatable/expandable balloon-like bio-polymers. The living core comprises the cell-matrix construct comprised of HDFs, for example, seeded in a scaffold. The method comprises surgically removing a damaged cartilage from a patient and inserting the hybrid construct into the cavity generated after the foregoing surgical intervention. The balloons of the inert structure are successively inflated within the target area, such as a joint, for example. Also disclosed herein are methods for growing and differentiating human fibroblasts into chondrocyte-like cells via mechanical strain.

摘要: This disclosure is directed to restoring joints by deploying a resilient interpositional arthroplasty implant. Such implants function to pad cartilage defects, cushion, and replace or restore the articular surface, which may preserve joint integrity, reduce pain and improve function. The implant may endure variable joint compressive and shear forces and cyclic loads. The implant may repair, reconstruct, and regenerate joint anatomy, and thereby improve upon joint replacement alternatives. The walls of this invention may capture, distribute and hold living cells until aggregation and hyaline cartilage regrowth occurs. The implant may be deployed into debrided joint spaces, molding and conforming to surrounding structures with sufficient stability so as to enable immediate limb use after outpatient surgery. Appendages of the implant may repair or reconstruct tendons or ligaments, and menisci by interpositional inflatable or compliant polymer arthroplasties that promote anatomic joint motion.

摘要: Methods are disclosed for forming bone and/or cartilage in an avian subject. The methods include administering to the avian subject a therapeutically effective amount of a composition comprising avian mesenchymal stem cells and a hydrogel that supports the differentiation of the avian mesenchymal stem cells into cells of an osteogenic and/or condrogenic lineage. In some embodiments, methods are disclosed for repairing a bone defect and preventing infection, such as that associated bone fracture, in an avian subject. The methods include administering locally to the bone defect a composition comprising a therapeutically effective amount of avian mesenchymal stem cells and a hydrogel, such as a methacrylated gelatin hydrogel.

摘要: Disclosed is a cell which enables the reproduction of a cartilage tissue and has a proliferative ability. Also disclosed is a technique for providing a cell supply source which can be used in a definitive treatment of osteochondrosis deformans. A chondrocyte-like cell which has the same properties as those of a chondrocyte and can proliferate can be produced by selecting a combination of an Myc family gene and/or a Klf family gene and a SOX9 gene and introducing the combination into a somatic cell. The chondrocyte-like cell can be used for a medical purpose of cartilage regeneration.
