摘要: A device and a method are disclosed for collecting and culturing fish embryos and evaluating combined toxicity of thiamethoxam and tetraconazole. An embryo collection component and an embryo culture component are provided on a base plate. The embryo collection component includes a collection tray detachably connected to a bottom of a culture tube. The embryo culture component includes a cavity plate with a stopper inside and a top plate provided with a culture well. A bottom of the culture well is provided with a leakage hole; and after the top plate is installed into the cavity plate, the stopper occludes the leakage hole. The new device is used to carry out the combined toxicity effect test of thiamethoxam and tetraconazole on zebrafish embryos, which can be used to avoid development of pesticide mixtures that have a good preventive effect but pose increased toxicity risk to the ecological environment.

摘要: Methods for the production of reproductively sterile fish and aquatic animals for aquaculture, the aquarium trade, and control of invasive species are described. The methods include disruption of gonadal development through the administration of compounds that lead to the failure of fertile gonadal development. Compounds may be delivered to the eggs prior to fertilization or water activation or post fertilization and water activation by contacting unfertilized or pre-water-activated fertilized eggs or fertilized eggs in an immersion medium including the compound of interest. Compounds may be conjugated with a molecular transporter compound effective for chorionic transport of the conjugate. The compounds may be antisense Morpholino oligomers that are capable of effectively suppressing the expression of the dead end gene or other essential genes for germ cell development in fish and other egg-producing aquatic animals.

The invention is based upon identification of a novel and surprising catalytic activity for the family of TET proteins, namely TET1, TET2, TET3, and CXXC4. The novel activity is related to the enzymes being capable of converting the cytosine nucleotide 5-methylcytosine into 5-hydroxymethylcytosine by hydroxylation.

摘要: A method for producing a cell in which a target gene is knocked out, the method including the step of: introducing a CRISPR-Cas system into a cell having one or more kinds of target genes, the CRISPR-Cas system being able to produce (i) three or more kinds of guide RNAs for each of the one or more kinds of target genes and (ii) a Cas protein.
