Abstract: The present technology is directed to methods of preparing affinity chromatographic columns using streptavidin-conjugated particles and biotinylated affinity agents (i.e., biotinylated oligonucleotides, biotinylated antibodies or biotinylated antigen-binding fragments thereof). The methods of the present technology can be utilized to perform affinity capture assays in a high-throughput, efficient manner. Further, the affinity chromatographic columns disclosed herein are customizable due to the ability to utilize any biotinylated affinity agent. The methods herein utilize an on-line loading (i.e., loading the column with biotinylated affinity agent via a liquid chromatography system).

Abstract: An analysis method for a target substance includes: mixing a conjugate and a sample including the target substance to produce a mixture, the conjugate including a labeled substance and a first binding substance having activity to bind to the target substance; bringing the mixture into contact with a second binding substance fixed to a base; and detecting a phenomenon caused by the labeled substance bound to the target substance that is bound to the second binding substance. The first binding substance is a substance including a nucleic acid, or a substance including an amino acid. The base is provided in each of regions. The target substance to which the second binding substance binds is different among the regions.

Abstract: Systems and methods are described herein for a diluent-containing tube comprising a beveled grinding surface and operable to receive a plunger comprising a complementary beveled grinding surface. Solid matrices placed within the diluent can be ground between the grinding surfaces to release molecules into the diluent for detection. The plunger may further comprise a groove containing a test strip operable to be deployed into the diluent after grinding for detection of the released molecules. In certain embodiments, the test strip may be a lateral flow immunoassay rapid antigen detection (LFA) test strip. The solid matrix may be a tick and the molecules may be antigens or pathogens related to tick-borne illnesses.

Abstract: The present disclosure describes improved SELEX methods for producing aptamers that are capable of binding to target molecules and improved photoSELEX methods for producing photoreactive aptamers that are capable of both binding and covalently crosslinking to target molecules. Specifically, the present disclosure describes methods for producing aptamers and photoaptamers having slower dissociation rate constants than are obtained using prior SELEX and photoSELEX methods. The disclosure further describes aptamers and photoaptamers having slower dissociation rate constants than those obtained using prior methods. In addition, the disclosure describes aptamer constructs that include a variety of functionalities, including a cleavable element, a detection element, and a capture or immobilization element.
