摘要: Provided herein are methods and compositions relating, in part, to the generation of human progenitor cells committed to the lung lineage and uses of such cells for treatment of lung diseases/disorders or injury to the lung. Whether an adult stem cell can be isolated from human adult lung remains controversial in the art and at present, methods for isolating and using adult lung stem cells from humans lack reproducibility. Thus, the methods and compositions described herein are advantageous over the present state of knowledge in the art and permit the generation of human lung progenitor cells for treatment, tissue engineering, and screening assays.

摘要: The present disclosure provides a composite decellularized matrix membrane and use thereof. The composite decellularized matrix membrane prepared by the present disclosure is prepared by wrapping one layer of a polymer membrane with two layers of decellularized matrix membranes. The decellularized matrix membrane is prepared from at least a biomaterial of a porcine small intestine, a porcine bladder, and porcine skin. The polymer membrane is prepared from at least a polymer material of polycaprolactone, polydimethylsiloxane, polyurethane, polylactic acid-glycolic acid, polyvinyl alcohol, and polyhydroxy fatty acid ester. The composite decellularized matrix membrane prepared by the present disclosure can prevent calculi, makes a patient suffer less, recover faster, live better, and cost less, while medical resources are saved, and hope for treatment is also brought for some patients with a contraindication to an in-situ ileal neobladder surgery.

摘要: The present disclosure provides methods for forming stable three-dimensional vascular structures, such as blood vessels and uses thereof. More specifically, the present disclosure provides methods for culturing differentiated endothelial cells that include an exogenous nucleic acid encoding ETV2 transcription factor on a matrix under conditions that express exogenous ETV2 protein in the endothelial cell to form stable three-dimensional artificial blood vessels without the use of a scaffold, pericytes or perfusion. The present disclosure also provides stable three-dimensional blood vessels that are capable of autonomously forming a functional three-dimensional vascular network, and uses thereof. In addition, the present disclosure includes methods for vascularizing an organoid and a decellularized organ by culturing the organoid or decellularized organ with endothelial cells that include an exogenous nucleic acid encoding ETV2 transcription factor under conditions that express exogenous ETV2 protein in the endothelial cell to vascularize the organoid or decellularized organ.

摘要: Intravesical devices are provided that are wholly deployable within the bladder of a patient in need of treatment and are well tolerated by the patient. The device may include an elastic body having a retention shape having (i) dimensions that provide intravesical mobility and that prevent voiding of the medical device through the urethra, and (ii) dimensions, buoyancy, or both, that exclude the medical device from entering the orifices of the ureters. The elastic body may exert a maximum acting force less than 1N when compressed to a shape with a maximum dimension in any dimension of 3 cm. The device may include a drug for controlled release within the bladder, for treatment of the bladder or a regional tissue. Methods of treatment are also provided that include selecting a patient in need of treatment in the bladder where tolerability of the treatment is a primary concern.

摘要: An epithelial cell tube for tracheal transplantation according to an embodiment includes a columnar support, and a whole layer of epithelial cells positioned on an outer peripheral surface of the columnar support. The epithelial cells are oriented so that cilia of the epithelial cells face a lumen side. The epithelial cell tube can be applied to an injured portion to provide an epithelial cell layer which is engrafted in tissues and can induce reduction of stenosis of trachea and inflammation.

摘要: Described herein are tissue grafts derived from the placenta that possess good adhesion to biological tissues and are useful in would healing applications. In one aspect, the tissue graft includes (1) two or more layers of amnion, wherein at least one layer of amnion is cross-linked, (2) two or more layers of chorion, wherein at least one layer of amnion is cross-linked, or (3) one or more layers of amnion and chorion, wherein at least one layer of amnion and/or chorion is cross-linked. In another aspect, the grafts are composed of amnion and chorion cross-linked with one another. In a further aspect, the grafts have one or more layers sandwiched between the amnion and chorion membranes. The amnion and/or the chorion are treated with a cross-linking agent prior to the formation of the graft. The presence of the cross-linking agent present on the graft also enhances adhesion to the biological tissue of interest. Also described herein are methods for making and using the tissue grafts.
