Abstract: The disclosure relates to methods for producing mesodermal lineage cells, cardiac lineage cells, hematopoietic lineage cells, retinal lineage cells, and combinations thereof. In some aspects, the disclosure relates to methods of producing mixed tissue organoids comprising retinal lineage cells and cardiac lineage cells. When interaction of WDR5 protein at the binding pocket/interaction surface with RBBP5/MYC/KANSL2 is disrupted in the embryonic stem cell for a set period of time after differentiation of the embryonic stem cell has begun, and the disruption of that interaction is subsequently removed, differentiation of the embryonic stem cell to a mesodermal lineage cell is obtained. Such mesodermal lineage cells may, in some methods of the disclosure, be subsequently differentiated into cardiac lineage cells and hematopoietic lineage cells. The disclosure also relates to method for producing mixed lineage organoids, comprising retinal and mesodermal, including cardiac, lineage cells in a single organoid. The disclosure also relates to cells and organoids obtained by the methods, uses of the cells and organoids, and kits comprising the cells and/or reagents for producing them.

Abstract: Provided are chemical inducers of lineage reprogramming (CiLR) which include glycogen synthase kinase inhibitors, TGFβ receptor inhibitors, cyclic AMP agonists or histone acetylators. Also provided is a method of inducing lineage reprograming in a partially or completely differentiated cell of a first type into a cell with characteristics of a second and different lineage. The method includes: contacting a cell with the CiLR for a sufficient period of time to result in reprograming the cell into a modified XEN-like cell which is subsequently programmed into a cell with characteristics of a second and different lineage.

Abstract: Methods for generating human midbrain immature neurons and mature dopaminergic neurons from neural progenitor cells are provided. The immature and mature midbrain neurons are obtained from neural progenitors such as committed midbrain neural stem cells (NSCs) and midbrain neural progenitor cells (midbrain NPCs), which themselves are generated from human pluripotent stem cells. The methods for generating midbrain immature and mature neurons use chemically-defined culture media that allow for generation of mature dopaminergic neurons in as little as 23 days. Culture media, isolated cell populations and kits are also provided.

Abstract: The present invention relates to 3D brain organoids, uses thereof, methods and culture medium for generating such organoids. An aspect of the invention provides brain organoids and methods of generating such organoids with bilaterally symmetric optic vesicles, containing both neuronal and non-neuronal cell types, and exhibiting functional circuitry. These organoids can be generated within short time intervals (e.g., 50 days) and therefore are useful for medical modelling and applications.
