Disclosed are disubstituted benzidine derivatives having on both the 3- and 3''-position a substituent W-X-A-On-, subscript n is zero or one, A is a divalent lower alkylene chain with from about 2 to about 7 carbons, X is the divalent carbonyloxy or sulfonyloxy group, W is hydrogen or other chromogen-reactiveindicator compatible cation; and the chromogen-reactive-indicator compatible acid addition salts of those benzidine derivatives. Also disclosed is the improvement in an analytical test, wherein is used a color change reagent preparation comprising peroxidase, an oxygen-oxidoreductase specific to a particular substance to be tested and for which there exists a specific oxygenoxidoreductase, and a chromogen-reactive-indicator which is one of the foregoing disubstituted benzidine derivatives or a chromogen-reactive-indicator compatible acid addition salt of it.

A diagnostic composition is prepared which contains as a chromogen a diamino-biphenyl derivative in which at least one of the hydrogen atoms in the amino groups thereof is replaced by a lower alkyl group by reaction with an alkylating agent. A diagnosing instrument is prepared by impregnating a bibulous, porous support in a solution comprising the diagnostic composition and optionally a saccharide oxidase, peroxidase and a guaiac, and, if necessary, a buffer, a surfactant, a background adjusting dye and/or an agent for increasing the viscosity of the composition. This diagnosing instrument can be handled in a simple and convenient way and is used to identify semi-quantitatively a saccharide in body fluids or excrements of humans or animals with reliability.

The present invention provides an improved method for staining slides using immunochemical reagents. The method comprises the following steps. The assay region of a slide (the region containing the tissue section)is washed with an improved rinsing solution comprising water and a detergent. An evaporation inhibitor liquid is applied to the slide to cover the assay region. For antigens requiring unmasking, the tissue section is combined with an improved, stabilized proteolytic enzyme solution. The slide is rinsed, and the evaporation inhibitor liquid is reapplied to the slide. A primary antibody in an improved diluent containing globulins from the same species as a second antibody is combined with the tissue section for a time sufficient for substantially complete antibody binding. The slide is rinsed, and the evaporation inhibitor liquid is reapplied. A labeled second antibody in the improved diluent is combined with the tissue section for a time sufficient for substantially complete antibody binding. The slide is rinsed, and the evaporation inhibitor liquid is reapplied to the slide. Color development reagents, including a stabilized peroxidase chromophore formulation, are combined with the tissue section for a time sufficient for color development. The stabilized peroxidase chromophore formulation consists essentially of a peroxidase chromophore, at a concentration in the working range of the enzyme, an acidic buffer, a reducing agent, and a glycol. Preferred chromophores include 3,3'-diaminobenzidine and tetrahydrochloride (DAB) and 3-amino-9-ethylcarbazole (AEC). After color development, the tissue section is washed and is then ready for analysis.

The disclosure is directed to compositions, kits, and methods for performing colorimetric analysis. A composition for a colorimetric assay can be prepared that includes an aromatic amine chromogenic substrate and a zwitterionic solubilization reagent that is N,N,N-trimethylglycine (betaine). The zwitterionic solubilization reagent is highly useful as a non-encapsulating solubilization reagent and provided faster kinetics and increased dye content of an aromatic amine chromogenic substrate in an enzymatic reaction. In turn, this can result in improvements in detection of an analyte in a sample and also can increase the sample throughput. The aromatic amine chromogenic substrate and N,N,N-trimethylglycine solubilization reagent can be used in assays such as ELISAs in order to provide a more accurate and faster detection of analytes in a biological sample.

Further disclosed are chromogen-reactive indicator compositions having as their chromogen constituent one of the foregoing benzidine derivatives. These compositions (1) are useful to test for a substance for which there exists a specific oxygen-oxidoreductase, such as a sugar, an amino acid, uric acid, and the like, and thus (2) contain also a peroxidase, a hydrogen-peroxide-oxidoreductase, and a specific oxygen-oxidoreductase for the specific material in the testing of which the composition is to be used.
