Abstract: Provided herein are methods of reducing bioburden of (e.g., sterilizing) a chromatography resin that include exposing a container including a composition including a chromatography resin and at least one antioxidant agent and/or chelator to a dose of gamma-irradiation sufficient to reduce the bioburden of the container and the chromatography resin, where the at least one antioxidant agent and/or chelator are present in an amount sufficient to ameliorate the loss of binding capacity of the chromatography resin after/upon exposure to the dose of gamma-irradiation. Also provided are reduced bioburden chromatography columns including the reduced bioburden chromatography resin, compositions including a chromatography resin and at least one chelator and/or antioxidant agent, methods of performing reduced bioburden column chromatography using one of these reduced bioburden chromatography columns, and integrated, closed, and continuous processes for reduced bioburden manufacturing of a purified recombinant protein.

Abstract: An apparatus for extracting an analyte from a liquid sample having a container with an entrance, an exit, and a passage therebetween for passage of a liquid sample containing an analyte, the container having a full diameter bed region and a reduced diameter bed region. The container includes a layered construction extending across the passage, having from top to bottom one or more of: (i) an upper flow distributor/support layer, (ii) an upper compression layer, (iii) an extraction layer of microparticulate extraction medium adjacent to the layer (ii), and (iv) a lower compression layer located adjacent to the extraction layer (iii), optionally including one or more air gap layers. At least some of the layers are located in the full diameter bed region, and some of the layers are located in the reduced diameter bed region. The apparatus may have a plurality of containers arranged in an array and/or in series so as to provide multi-stage filtration or extraction.

Abstract: The invention provides a packing method for high efficiency chromatography columns starting from dry swellable particles, as well as columns packed by the method and the use of the columns in separation of biomolecules. In the packing method, an amount of dry swellable particles sufficient to give a swollen volume in a liquid of about 105-120% of the column chamber volume is transferred to the column, the column is closed and the liquid is provided to the column.

Abstract: A chromatography column (2) containing a bed of packed particles (22, 24, 26, 28, 30), wherein the packed particles comprise fused core particles and the particle diameters of the packed particles vary along the column. Preferably, the particles (2, 24, 26, 28, 30) are arranged according to their average particle diameter, in order of increasing average particle diameter from the inlet end (4) to the outlet end (6). The bed may comprise a plurality of bed sections and each bed section has an average particle diameter calculated from the particles in that section and there are at least two different average particle diameter bed sections, wherein the particles of each bed are separated from particles of an adjacent bed by a partition that is liquid permeable to allow through a flow of mobile phase. A high column efficiency can be provided with lower pressure drop per unit length of the column.
