The detector (3) has a detection surface (3a) which is divided into multiple detection regions (4a). The detector is additionally designed to derive (S1) detection signals (4c) of individual detection regions (4a) with respect to time, subsequently rectify (S2) the signals, preferably by generating an absolute value or squaring, and summing or averaging (S3) the derived and rectified detection signals of all of the detection regions and then providing same as an output signal (6c). Laser speckle microrheology is used to determine a mechanical property of a biological tissue, namely, an elastic modulus. Speckle frames may be acquired by illuminating a coherent light and capturing back-scattered rays in parallel and cross-polarized states with respect to illumination. The speckle frames may be analyzed temporally to obtain diffuse reflectance profiles (DRPs) for the parallel-polarized and cross-polarized states. A scattering characteristic of particles in the biological tissue may be determined based on the DRPs, and a displacement characteristic may be determined based at least in part on a speckle intensity autocorrelation function and the scattering characteristic. A size characteristic of scattering particles may be determined based on the DRP for the parallel polarization state. The mechanical property may be calculated using the displacement and size characteristics.

An algorithm for determining viscoelastic modulus of an optically scattering biofluid that takes into account variable scattering and/or absorption characteristics of the biofluid. A correction to mean square displacement value characterizing the Brownian motion of light scatterers is introduced based on a polarization-sensitive Monte-Carlo ray-tracing taking into account optical properties of the biofluid determined with the use of laser speckle rheology measurements. In contradistinction with a diffusion model, the correction-implemented determination of the viscoelastic modulus applies to a biofluid with substantially any concentration of light-scattering particles.
