    摘要: Novel anti-idiotype monoclonal antibodies are described which are capable of specifically reacting with the idiotype of human anti-gp120 antibodies, of inhibiting the binding between the gp120 antigen and human anti-gp120 antibodies, and of evoking a neutralising anti-gp120 immune response in an animal host to which they are administered. The anti-idiotype antibodies of the invention can be identified based on the amino acid sequences of the variable portions of their light and heavy chains. In addition, a method for obtaining a panel of anti-idiotype monoclonal antibodies, expression vectors and transformed host cells usable in a recombinant DNA procedure in order to generate the aforesaid anti-idiotype monoclonal antibodies, as well as the therapeutic, prophylactic and diagnostic use of such antibodies are disclosed.

    摘要: The invention relates to Ab2-type anti-idiotypic antibodies and fragments thereof which mimic HVI-1 epitopes that are otherwise cryptic to the immune system and which antibodies or fragments thereof are directed against potently neutralizing anti-HIV-1 antibodies. The invention further relates to a hybridoma cell line 3H6 expressing the anti-idiotypic antibody and to pharmaceutical compositions containing the antibody or fragment thereof. The invention also relates to HIV-1 neutralizing Ab3-type antibodies elicited upon administration of the Ab2-type anti-idiotypic antibody or fragment thereof and to pharmaceutical compositions containing them.     摘要: Nucleotide sequences encoding the variable heavy and light chains of the murine monoclonal antibody 1F7 are disclosed. The 1F7 antibody is an effective immune modulator that has anti-idiotypic binding affinity for anti-HIV antibodies. Polypeptides containing at least one complementarity-determining region (CDR) or framework-determining region (FR) of the variable heavy or variable light chains of 1F7, as well as the polynucleotides encoding them, can be used to modulate the immune response to HIV infection. Unnecessary, and potentially adverse, murine segments of the variable chains falling outside the CDRs can be replaced with human sequences to afford humanized chimeric antibodies and antibody fragments.

    摘要: The present invention provides a method of manufacture of an anti-paratopic antibody comprising the steps of: (1) selecting from a pool of antibodies occurring in one species a prototypic set the members of which are effective in binding a specific antigen (or antigen epitope), and (2) utilizing one or more members of said prototypic set, or paratopic fragments thereof, as an immunogen in a host of a different species, or in an in vitro incubation system comprising cells derived from the same or a different species, to produce antibodies having a characteristic which is anti-paratopic with respect to said immunogen to produce a synthetic replicate of the specific antigen or epitope. Antigen (or antigen epitope), and monoclonal antibodies, vaccines and processes of immunization employing the product of the method of manufacture are also described.
