The invention provides a novel, general, and facile strategy for the creation of small molecules with high structural and stereochemical complexity. Aspects of the methods include ring system distortion reactions that are systematically applied to rapidly convert readily available natural products to structurally complex compounds with diverse molecular architectures. Through evaluation of chemical properties including fraction of sp3 carbons, ClogP, and the number of stereogenic centers, these compounds are shown to be significantly more complex and diverse than those in standard screening collections. This approach is demonstrated with natural products (gibberellic acid, adrenosterone, and quinine) from three different structural classes, and methods are described for the application of this strategy to any suitable natural product.

The present invention relates to a method for the production of a diterpene or a glycosylated diterpene, which method comprises: a. fermenting a recombinant microorganism of the genus Yarrowia in a suitable fermentation medium at a temperature of about 29° C. or higher, wherein the microorganism comprises one or more nucleotide sequence(s) encoding: a polypeptide having ent-copalyl pyrophosphate synthase activity; a polypeptide having ent-Kaurene synthase activity; a polypeptide having ent-Kaurene oxidase activity; and a polypeptide having kaurenoic acid 13-hydroxylase activity and whereby expression of the nucleotide sequence(s) confer(s) on the microorganism the ability to produce at least steviol; and b. recovering the diterpene or glycosylated diterpene.

A cardiogenin major isomer is obtained from a methanol extract of Geum japonicum and separated from its minor isomer. The separation of the two isomers can be achieved by chiral phase chromatography, e.g., using a Chiralpak® IC™ column. The purity of the isolated cardiogenin major isomer can be further increased by crystallization, yielding isolated cardiogenin major isomer having HPLC purity as high as 98.97% (a/a) at 210 nm and a potency of 95.50%) (w/w).

Synthetic triterpenoids and methods of using the same to induce gene expression and differentiation of stem or progenitor cells are provided. Furthermore, the present invention provides methods for producing a cell, such as a stem or progenitor cell, with induced gene expression by contacting a stem or progenitor cell with an effective amount of a synthetic triterpenoid to induce the expression of one or more of SOX9 (Sex determining region Y-box 9), COL2A1 (Type II Collagen (alpha1)), TGF-βI, TGF-p2, TGF-33, BMP2, BMP4, BMPRII (Bone Morphogenic Protein Receptor II), SMAD (Small Mothers Against Decapentaplegic) 3, SMAD4, SMAD6, SMAD7, TIMP (Tissue Inhibitor of Metalloproteinase)-1 or TIMP-2 in the stem or progenitor cell, wherein the stem or progenitor cell is not a mesenchymal stem cell, periosteium cell or osteoprogenitor cell. In one embodiment, the stem or progenitor cell is multipotent.
