The instant invention involves the use of a combination of preparatory steps in conjunction with mass spectroscopy and time-of-flight detection procedures to maximize the diversity of biopolymers which are verifiable within a particular sample. The cohort of biopolymers verified within such a sample is then viewed with reference to their ability to evidence at least one particular disease state; thereby enabling a diagnostician to gain the ability to characterize either the presence or absence of said at least one disease state relative to recognition of the presence and/or the absence of said biopolymer, predict disease risk assessment, and develop therapeutic avenues against said disease.

A method is provided for covalently linking carbohydrates, proteins, nucleic acids, and other biomolecules under neutral conditions, using a Diels-Alder cycloaddition reaction. In an example, activated carbon-carbon double bonds were attached to free amino sites of a carrier protein, and a conjugated diene was attached to a carbohydrate hapten. Spontaneous coupling of the carbohydrate and the protein components under very mild conditions provided glycoconjugates containing up to 37 carbohydrate hapten units per carrier protein molecule. The method is also applicable to the immobilization of biomolecules on gel or solid supports. The conjugated products are useful as immunogens and as analytical and diagnostic reagents.

An immunological assay method is disclosed which utilizes recombinant antigen, rNcp29, derived from an immunodominant surface antigen of Neospora caninum tachyzoites. Specifically, an ELISA method is disclosed. The method provides sensitive and specific detection of antibodies in sera of infected animals and does not exhibit cross-reaction with antisera against related parasites such as T. gondii. The ELISA method is used to screen animals for the presence of serum antibodies specific to recombinant Ncp29.

This invention relates to monoclonal antibodies which bind to non-enhancing protective epitopes on serotype A, B, C and D strains of C. neoformans, such protective epitopes containing acetyl groups in the polysaccharide of the epitopes. Other monoclonal antibodies of this invention are serotype specific, and bind to acetyl groups on polysaccharide capsule protective epitopes of serotype D strain C. neoformans only. This invention further relates to methods for producing these monoclonal antibodies. These monoclonal antibodies may be passively administered to treat and prevent cryptococcal infection, such as Cryptococcal meningitis, in immunosuppressed patients. These monoclonal antibodies may also be used for detection of fungal infection, for the development of diagnostic serotyping of clinical isolates, and as therapeutic adjuncts to anti-fungal antibiotic therapy.

Transfusion of contaminated blood has become the major route of transmission for Chagas' disease. Current screening tests are insensitive and yield conflicting results, while confirmatory assays do not exist. The present invention relates to antigens and their use for serological diagnosis of Chagas' disease. More specifically, the present invention concerns assays which are able to reliably and accurately detect the presence of antibodies to various specific antigens of Trypanosoma cruzi in a highly sensitive and specific manner.
