    Abstract: An image acquisition method in which a pulsed illumination beam emitted from a light source is scanned while being focused at a sample, signal light generated as a result of a non-linear optical process at each scanning position is detected, and an image of the sample is generated on a basis of the detected signal light, the image acquisition method including: acquiring a mixed image, which includes in-focus signal light generated at a focal position of the illumination beam in the sample and which also includes out-of-focus signal light; acquiring an image of the out-of-focus signal light on a basis of a plurality of mixed images having mutually different intensities of the out-of-focus signal light; and acquiring an image of the in-focus signal light by subtracting the image of the out-of-focus signal light acquired, from the mixed image acquired.

    Abstract: An observation assistance device of the present invention includes: an information storage unit that stores sample state information showing a sample state change, a type of sample, a type of fluorescent substance, and an observation condition in fluorescence observation performed on a predetermined sample, and under a predetermined observation condition; an input unit via which inputs the type of sample to be observed and the type of fluorescent substance to be used; an assistance-display generating unit that reads sample state information and an observation condition of fluorescence observation performed on the same type of sample as the type of sample input, using the same type of fluorescent substance as the type of fluorescent substance, and generates an assistance display showing the sample state information in relation to the two or more parameters included in the observation condition; and a display unit that displays the generated assistance display.

    Abstract: A point-spread-function measurement device includes: a scanner that scans two illumination light beams emitted from a light source; an illumination optical system that radiates the two illumination light beams scanned by the scanner onto a sample; a relative-position adjustor that changes a relative irradiation position, in the sample, between the two illumination light beams radiated by the illumination optical system; a detection optical system that detects signal light generated at an overlapping position, in the sample, of the illumination light beams radiated by the illumination optical system; and a calculator that calculates a point spread function based on the signal light detected by the detection optical system and the relative irradiation position between the two illumination light beams when the signal light is detected.

    Abstract: Provided is a microscope provided with: a scanner that scans an excitation beam coming from a light source; an objective optical system that focuses the scanned excitation beam onto a sample and that collects fluorescence generated at individual scanning positions in the sample; a detector that detects the collected fluorescence; a light blocking member that is disposed between the detector and the system and that partially blocks the collected fluorescence; a switching portion that switches the positional relationship between the member and a light-focusing point of the excitation beam in the sample between an optically conjugate positional relationship, in which an in-focus fluorescence generated at the light-focusing point passes through the member, and a non-conjugate positional relationship, in which the in-focus fluorescence is blocked by the member; and a computing portion that computes a difference between fluorescence signals acquired by the detector in the two positional relationships.
