摘要: Methods of quantitating extra-cellular vesicle surface markers are provided. Aspects of the methods include comparing: a mean fluorescence intensity of a surface marker of interest (surface marker MFI) of a labeled extra-cellular vesicle (EV) sample with a calibration plot obtained from a liposome calibration composition to quantitate the surface marker on extra-cellular vesicles of the EV sample. Also provided are compositions that find use in practicing embodiments of the invention.

摘要: Methods of determining whether a sample includes an antibody that binds to a coronaviral antigen are provided. Aspects of the methods may include combining the sample, a mammalian cell displaying on a surface thereof an antigenic determinant of the coronaviral antigen that is specifically bound by the antibody, and a labeled secondary binding member that binds to the antibody to produce an assay composition; and flow cytometrically assaying the assay composition for the presence of a signal from the labeled secondary binding member to determine whether the sample includes the antibody. Also provided are methods of assessing a subject for an immune response to a coronaviral infection and methods of producing a mammalian cell displaying on a surface thereof an antigenic determinant of a coronaviral antigen. Also provided are systems, expression vectors, mammalian cells, and kits for practicing the subject methods.

摘要: Aspects of the present disclosure include methods for phase correcting signals from a light detection system (e.g., in a flow cytometer). Methods according to certain embodiments include detecting light from a sample having particles in a flow stream with a light detection system that includes a brightfield photodetector configured to generate a brightfield data signal and a fluorescence detector configured to generate a fluorescence data signal and calculating a phase correction for the fluorescence detector based on the relative phase between the brightfield data signal and the fluorescence data signal. Systems having a processor with memory operably coupled to the processor having instructions stored thereon, which when executed by the processor, cause the processor to calculate a phase correction for a fluorescence detector based on a brightfield data signal and a fluorescence data signal from the fluorescence detector are also described. Integrated circuit devices (e.g., field programmable gate arrays) having programming for practicing the subject methods are also provided.

摘要: The present invention provides labeling and capture reagents that comprise phosphorothioate oligonucleotides (PS-ODN). The PS-ODN bind to all white blood cells (leukocytes) in an indiscriminative fashion and enable the labeling, capture, or concentration of leukocytes in a manner that preserves the antigenic integrity of the cells. Methods for the use of phosphorothioate oligonucleotides are provided.

摘要: Drug-loaded microbead compositions including a plurality of individual microbeads of a biodegradable material, a plurality of individual vesicular agents, a first therapeutic agent, and a second therapeutic agent different from the first therapeutic agent. The vesicular agents include at least one lipid bilayer surrounding a vesicular core. The first therapeutic agent is associated with the individual vesicular agents. For example, the first therapeutic agent may be contained within the individual vesicular agents or may be associated with the individual vesicular agents by ionic or non-covalent interaction. The second therapeutic agent is different from the first therapeutic agent and contained within the individual microbeads or associated with the individual microbeads through ionic or non-covalent interaction. The second therapeutic agent may or may not be associated with the individual vesicular agents. Drug-loaded biodegradable microbead compositions include microbeads of biodegradable material and a therapeutic agent associated with N the microbeads.
