    摘要: The present invention relates to the identification of novel metallo-proteases (MP) in Gram-positive microorganisms. The present invention provides the nucleic acid and amino acid sequences for Bacillus MP. The present invention also provides host cells having a mutation or deletion of part or all of the gene encoding MP. The present invention also provides host cells further comprising nucleic acid encoding desired heterologous proteins such as enzymes. The present invention also provides cleaning compositions comprising an MP of the present invention.

    摘要: The present invention relates to methods for making recombinant thermophilic bacteria of the family Bacillaceae. In particular, recombinant thermophilic Bacillaceae are provided which have been engineered to produce butanol and/or butyrate. Preferably, heterologous nucleic acid molecules encoding one or more butanol or butyrate biosynthetic pathway enzymes are introduced into a thermophilic Bacillaceae in order to produce a recombinant thermophilic Bacillaceae which is capable of producing butanol. The Bacillaceae is preferably of the genus Geobacillus or Ureibacillus. The invention also relates to a method of producing butanol using the Bacillaceae of the invention.

    摘要: Variants of B. licheniformis alpha-amylase advantageously exhibit improved enzymatic performance. Suitable variants include those with an altered charge distribution on the surface of the enzyme or with altered active site residues. Structural modeling can inform the choice of amino acid modifications so that modified amino acids correspond to residues found in more active alpha amylases, for example. Compositions comprising the variants are useful in methods of cleaning surfaces, laundering textiles, desizing, treating starch, e.g., liquefaction and saccharification, and hydrolyzing biofilms off various substrates.

The protease prodomain of the present invention is fused to a second protein to form a protease prodomain fusion protein. The presence of a protease prodomain protein in a fusion protein allows for easy and selective purification of the second protein by incubation with the corresponding protease.
