摘要: In certain example embodiments, the invention provides a method of generating an ex vivo cell-based system comprising dissociating an original tissue sample obtained from a subject into a single cell population; determining an in vivo phenotype of the tissue sample by conducting single-cell RNA analysis on a first portion of the single cells; establishing an ex vivo cell-based system from a second portion of the single cells; and culturing the ex vivo cell-based system in a medium or conditions selected to maintain the in vivo phenotype. In some embodiments, the original tissue sample is a tumor tissue sample, such as a pancreatic ductal adenocarcinoma (PDAC) tumor sample.

摘要: Provided is a culture medium containing amphiregulin for culturing primary breast epithelial cells and a culture method involving using the medium. In the culturing method, primary cells are cultured on a culture vessel coated with an extracellular matrix gel by using the culture medium, and the primary cells grow on the culture vessel, which has been coated with the extracellular matrix gel, and proliferate rapidly under the combined action of nutrient factors and an extracellular matrix contained in the primary cell culture medium. The cell model obtained by the primary cell culture medium and the primary cell culture method can be used for the evaluating the efficacy of and screening drugs.

摘要: The present invention is directed to a recombinant herpesvirus comprising a heterologous peptide and optionally polypeptide ligand capable of binding to (a) target molecule(s) and fused to or inserted into glycoprotein D. The recombinant herpesvirus may additionally comprise modifications for detargeting the virus from the natural receptors of gD. This allows the herpesvirus to efficiently target a cell for therapeutic purposes and a cell for virus production. The present invention further comprises a pharmaceutical composition comprising the herpesvirus, the herpesvirus for use in the treatment of a tumor, infection, degenerative disorder or senescence-associated disease, a nucleic acid and a vector coding for the gD, a polypeptide comprising the gD, and a cell comprising the herpesvirus, nucleic acid, vector or polypeptide. Moreover, a method for infecting a cell with the herpesvirus or for producing the herpesvirus is disclosed.

The core of the technology is as follows: (1) solid tumor of lung cancer are treated by using a mild cell dissociating reagent, and lung cancer cells in pleural effusion are dissociated by a mild method, ensuring the vitality of cancer cells to the greatest extent; (2) a special serum-free medium is prepared, and tumor cells derived from solid tumor tissues of lung cancer are cultured in vitro by using a suspension culture system, ensuring the normal amplification of the cancer cells while eliminating the interference of normal cells to the greatest extent.

摘要: Provided herein are methods for producing exosomes that contain RNA transcribed from a specific mutant gene or a transgene. In one embodiment, the method comprises the steps of: generating a cell comprising a mutation of a gene by using a site-specific nuclease; culturing the cell in a medium that allows the cell to secrete to the medium an exosome containing an RNA transcribed from the gene and comprising the mutation; and collecting the medium that contains the exosome. The exosomes generated can be used as reference material or therapeutic delivery device.
