    摘要: In vitro delivery of the diphtheria toxin (DT) catalytic (C) domain from the lumen of purified early endosomes to the external milieu requires the addition of both ATP and a cytosolic translocation factor (CTF) complex. The results presented here demonstrate that β-COP plays an essential role in the cytosolic release of the C-domain and is mediated by a consensus peptide sequence found on several bacterial toxins and in HIV-1 reverse transcriptase. The invention features methods for inhibiting cell death that include the administration of compounds based on this consensus sequence that inhibit the translocation of the catalytic domain of toxins or transcription factors. Also featured are methods for identifying compounds that inhibit cell death, and methods for identifying compounds that promote cell death by blocking or accelerating, respectively, the rate of toxin/factor endosomal translocation.

    摘要: Disclosed is a method for identifying activators of a transition metal-dependent repressor of virulence gene expression in infectious prokaryotic pathogens. The method utilizes genetic circuitry that represents the response of a given prokaryote to nutritional stress and the expression of genes that contribute to the establishment of the infectious process. The exposure of recombinant cells or a cell-free system containing the genetic circuitry to a non-metal ion test substance that activates the repressor produces a detectable response. The method is applicable for any prokaryote employing metal ion-dependent repressors to regulate specific gene expression, specifically as it pertains to virulence determinant expression.

    摘要: Apparatus for use in gyroscopically ejecting a spacecraft supported in the open end of a spacecraft supporting cradle. The invention comprises a separable ejection restraint mechanism attached between the cradle and the spacecraft. The mechanism restrains the thrust force applied to the spacecraft for a predetermined time period in order to dissipate undetermined forces acting upon the spacecraft prior to the ejection thereof. The mechanism comprises a separable resilient portion and a releasable clamp portion coupled thereto. The resilient portion comprises a spring loaded anchor rod arrangement while the releasable clamp portion comprises a spring loaded split ball clamp arrangement. The invention also contemplates the use of a flexible carrier coupled between the spacecraft and the cradle. The invention acts as a tether connected between the cradle and spacecraft which acts to establish a determinant state of force on the cargo and eliminate unwanted ejection-related problems prior to cargo deployment. In addition, the release of a single tether employed as the sole cargo release initiator minimizes the possible imparting of uncontrolled momentum to the cargo due to structural deformation energy.

    摘要: An air-operated engine system usable for driving a vehicle by means of compressed air from a rechargeable storage tank. The engine of the system has cylinders containing driving pistons connected to a crankshaft. Compressed air from the storage tank is supplied at regulated pressure to the cylinders for power strokes by means of intake valves and the air is exhausted from the cylinders at the ends of the power strokes by means of exhaust valves. The intake and exhaust valves are operated by dual lobe cams on a camshaft driven from the crankshaft. The air is compressed by a hydrovane compressor which has a constant flow of air operating under 100 pounds pressure with control valves which will take in 25 pounds per minute up to 200 pounds. The output pressure is controlled by the input pressure.
