The objective of the present invention is to produce a novel (R)-2,3-butanediol dehydrogenase useful for the production of ketones, alcohols, and particularly optically active vicinal diols. It has been found that Pichia angusta produces novel (R)-2,3-butanediol dehydrogenase that shows the high activity and high stereoselectivity. Further, polynucleotide encoding this (R)-2,3-butanediol dehydrogenase was cloned, and the nucleotide sequence thereof was determined. The expression of the glycerol dehydrogenase was carried out in heterologous microorganisms.

The invention also provides antisense nucleic acid molecules, recombinant expression vectors containing DHDR nucleic acid molecules, host cells into which the expression vectors have been introduced, and nonhuman transgenic animals in which a DHDR gene has been introduced or disrupted. The invention still further provides isolated DHDR proteins, fusion proteins, antigenic peptides and anti-DHDR antibodies. Other trans-retinol dehydrogenases are also disclosed. Nucleic acid molecules which code for the proteins are also disclosed.

This invention provides (R)-2-octanol dehydrogenase that catalyzes oxidation-reduction reaction using NAD+ (NADH) as a coenzyme and the genes that encodes them. The enzymes of this invention can be obtained from microorganisms such as the genera Pichia, Candida, and Ogataea, and so on. It is possible to produce alcohols, in particular, alcohols such as (S)-4-halo-3-hydroxybutyric acid esters and (R)-propoxybenzene derivatives by reducing ketones with this (R)-2-octanol dehydrogenase. Moreover, the (R)-2-octanol dehydrogenase of this invention is excellent in activity and stereoselectivity.
