摘要: The present disclosure provides a method for separating SSCs from a frozen testicular tissue, and belongs to the field of cell biology. In the present disclosure, the method includes the following steps: resuscitating a frozen testicular tissue, digesting the resuscitated testicular tissue with a collagenase IV and a DNase I until seminiferous tubules are exposed and tube walls become coarse; washing the digested testicular tissue with a DPBS, conducting centrifugation and culture until cells migrate; collecting the migrated cells, and enriching and purifying SSCs. The method can maintain a milieu interieur of the SSCs to the greatest extent, and therefore cell viability, and less cell damages. Therefore, the method can be used for the separation of SSCs from frozen testicular tissues.

摘要: Mice are provided that comprise a reduction or deletion of ADAM6 activity from an endogenous ADAM6 locus, or that lack an endogenous locus encoding a mouse ADAM6 protein, wherein the mice comprise a sequence encoding an ADAM6 or ortholog or homolog or fragment thereof that is functional in a male mouse. In one embodiment, the sequence is an ectopic ADAM6 sequence or a sequence that confers upon a male mouse the ability to generate offspring by mating. Mice and cells with genetically modified immunoglobulin heavy chain loci that comprise an ectopic nucleotide sequence encoding a mouse ADAM6 or functional fragment or homolog or ortholog thereof are also provided.

摘要: There is described a method for extracting a target chemical compound from a cellular material in a sample. The method comprising the steps of: subjecting the sample to mechanical lysis to cause disruption of a cellular membrane in the cellular material; contacting the sample with an alkaline material to produce a lysate composition comprising the target chemical compound; and recovering the lysate composition from the sample. There is also described a method for producing a lysate composition comprising RNA from a mammalian bodily fluid sample comprising a cellular material. There is also described a method for extracting a nucleic acid from a cellular material in a bodily fluid or an inoculant derived therefrom.
