摘要: Disclosed in the present invention are an ionizable lipid molecule, a preparation method therefor, a composition containing same, an application thereof in the preparation of a vector for delivering a nucleic acid to a cell, and an application thereof in the preparation of a lipid nanoparticle (LNP). By improving the structure of the ionizable lipid molecule and adjusting an LNP component solution, the effect of an mRNA-LNP preparation is improved. The ionizable lipid molecule of the present invention has a structure of formula (I). The ionizable lipid molecule is synthesized with phospholipid, cholesterol, and polyethylene glycol by means of microfluidics to obtain the LNP, and the obtained LNP can improve the translation expression level of a load-mRNA in the cell, improve the effect of the mRNA-LNP preparation, and provide a theoretical basis for theoretical treatment of the personalized mRNA-LNP preparation.

摘要: Provided herein is a method for producing an ionizable lipid that comprises: (i) reacting fatty esters in a Claisen condensation reaction in the presence of a catalyst, the Claisen condensation employing a weak base and carried out at a temperature of between −10 and 60 degrees Celsius to produce a ketoester; (ii) reacting the ketoester produced in step (i) under conditions to produce a ketone from the ketoester in one or more steps via a hydrolysis and decarboxylation of the ketoester; and (iii) preparing the ionizable lipid from the ketone thereof using one or more synthesis steps resulting in an addition of an ionizable head group moiety to (a) the ketone; or (b) an alcohol produced from an optional reduction of the ketone to produce the alcohol, thereby producing the ionizable lipid. The ionizable lipid produced in step (iii) may be formulated in a drug delivery vehicle.

摘要: The present disclosure relates to the technical field of drug synthesis, and provides a method for preparing a key intermediate of ABT-737 and a method for preparing ABT-737. In the present disclosure, the compound having a structure represented by formula I is used as the starting material. The carboxyl group in the compound having a structure represented by formula I is first reduced to a hydroxyl group, followed by a vulcanization by a vulcanizing agent, and then an amination, a deprotection, a condensation, and a carbonyl group reduction, to obtain the key intermediate of ABT-737 having a structure represented by formula VI. Tert-butoxycarbonyl is used as the protecting group for the compound having a structure represented by formula I, and the subsequent intermediate containing the protecting group of tert-butoxycarbonyl is stable, and easy to be deprotected. This method is suitable for mass synthesis, with high product yield and low production cost, and thus has a good industrialization prospect.
