Abstract: A recombinant strain of F. diplosiphon was made by transforming wild type F. diplosiphon with a pGEM-7Zf (+) plasmid containing sterol desaturase gene (SD) via electroporation. The recombinant strain was designated B481-SD and overexpressed the sterol desaturase gene to result in enhanced lipid production. Selection made on NaCl enabled growth of the transformant to thrive up to 50 g L−1 NaCl. This strain was designated B481-SDH.

Abstract: Methods and compositions for increasing lipid production in cyanobacteria where the cyanobacteria is incubated in the presence of non-lethal amounts of antibiotic. Non-lethal doses of antibiotic weakens the cell membrane facilitating migration of fatty acids from the cell, reducing inhibition feedback of fatty acid precursor enzymes, leading to an increase in fatty acid production.

Abstract: A method for improving aquaculture growth and/or survival rate and hemocyte viability, comprising growing the aquaculture in the presence of cyanobacteria F. diplosiphon. The aquaculture may be a shellfish aquaculture, for example, shrimp, crayfish, oysters, clams, mussels, and crabs or combinations thereof. The aquaculture may be freshwater aquaculture, brackish aquaculture, or saltwater aquaculture. The F. diplosiphon may be a wild-type or an engineered/recombinant strain. The F. diplosiphon may be preferably added to aquacultures in amounts of 1 million colony forming units or more per ml of aquaculture volume.

Abstract: Zero-valent iron nanoparticle-mediated oxidative stress was evaluated in F. diplosiphon as a measure of lipid peroxidation using malondialdehyde and thiobarbituric acid-reactivity (MDA-TBA) assay and the 2′,7′-dichlorodihydrofluorescein diacetate (DCFH-DA) fluorogenic probe. Enzymatic response of nZVI-treated cultures was determined as a measure of FeSOD (iron superoxide dismutase) using western blotting and immunodetection. In addition, the distribution and surface assimilation of nZVIs was detected using transmission electron microscopy (TEM) and energy-dispersive X-ray spectroscopy (EDS).
