摘要:
The present invention concerns to recombinant influenza viruses and modified Vaccinia Ankara viruses (MVA), and to a process for construction of recombinant influenza viruses and modified vaccinia Ankara viruses (MVA) with genes that encode for the T. gondii parasite SAGI (MVA) and SAG2 (MVA and influenza) proteins, by means of a homologous recombination technique between two transfer vectors (for construction of MVA virus) and reverse genetics (for construction of influenza virus). Additionally, the present invention describes a vaccine composition using recombinant influenza viruses and modified vaccinia Ankara viruses (MVA), or recombinant adenoviruses and modified vaccinia Ankara viruses (MVA), for immunization against infections caused by the T. gondii parasite.

The present invention provides a novel use of coccidian, specifically relates to the use of coccidian as a vaccine live vector. The present invention further provides a live vaccine with coccidian as a vector, which is transgenic coccidian capable of expressing exogenous protein or stably transfected coccidian that contain expression vector and can express exogenous coccidian. The present coccidian vector live vaccine can induce organisms to simultaneously generate protective humoral and cellular immune responses (including the mucosal immune response), as well as generate memory responses, which can be readily carried out and has stable effect and high biological safety without generating immune tolerance.

The present invention provides compositions and methods for the prevention and treatment of primary and metastatic neoplastic diseases and infectious diseases, for stimulating an immune response in a subject, and for use as an alternative to interleukin-12 (IL-12) treatment. In particular, the present invention provides Apicomplexa-related proteins (ARPs) that have immune stimulatory activity and thus have uses in the treatment and prevention of cancer and infectious diseases and in immune modulation. Compositions comprising an ARP are provided. Methods of use of an ARP for the prevention and/or treatment of cancer and/or infectious diseases, for use as an alternative to interleukin-12 (IL-12) treatment, and for eliciting an immune response in a subject, are also provided.

Uracil auxotroph mutants of apicomplexans are provided which lack a functional carbamoyl phosphate synthase II (CPSII) enzyme. Also provided are T. gondii autoxtroph mutants which express exogenous antigens, and methods of protecting an animal against a T. gondii and non-T. gondii disease.

Described are coccidia strains and microbiological cultures comprising such strains. The coccidia strains are selected for tolerance to one group of coccidiostatic drugs, sensitivity to a second group of coccidiostatic drugs, and decreased proliferative capacity. Also described are compositions and vaccines based on such coccidia strain.

This invention relates to novel Eimeria proteins with immunogenic properties as well as to DNA sequences encoding these proteins. These proteins can be administered to poultry thereby protecting the birds against coccidiosis. In addition the DNA encoding these proteins can be used for the preparation of a vector vaccine against coccidiosis.

摘要:
A method for obtaining an immunogenic strain useful for producing a vaccine against Coccidiosis comprises the cycle of infecting at least one first group of specific pathogen-free donor birds with oocysts from an Eimeria species. Blood is then collected from these donor birds, and is then used to infect a second group of specific pathogen-free birds. Oocysts are collected from the second group of birds. These oocysts are then multiplicated to complete the cycle. The cycle is then repeated using the multiplicated oocysts. After a total of about three cycles, a final antigen may be harvested and utilized as a source to generate oocysts for a vaccine.

A vaccine for in ovo vaccination against avian coccidiosis produced by a method including obtaining the coccidial oocysts from a fecal suspension, homogenizing the fecal suspension, separating the oocysts from the fecal debris by either salt flotation using sodium sulfate or gas flotation using air, sporulating the oocysts using hydrogen peroxide and air sparging, bleaching the sporulated oocysts, washing the bleached oocysts, concentrating the sterile washed oocysts and combining the concentrates of various species of coccidial oocysts, and producing a vaccine. The method in whole or in part can be applied to other kinds of encysted protozoa to produce vaccines for various types of animals.

The invention provides polypeptide fragments derived from TgAMA-1, nucleic acids that encode the polypeptide fragments, and TgAMA-binding polypeptides such as antibodies. Methods for using the polypeptide and nucleic acid molecules to produce vaccines are also provided. In addition the invention provides methods involving use of the polypeptides, nucleic acids, and binding polypeptides, such as antibodies, for the prevention and treatment of Toxoplasmosis.
