To provide a method of efficiently removing excitation light in a device for measuring fluorescence emitted from samples on a measuring surface of a substrate while illuminating the samples with excitation light. The method is a fluorometry characterized in that an excitation light illumination portion where the samples are illuminated with the excitation light and a light detecting portion where measurements are made of the fluorescence are placed in such a manner as to make it possible to prevent the excitation light from approaching the light detecting portion, and measurements of the fluorescence emitted from the samples on the measuring surface of the substrate are made in such a manner as to relatively move the samples from the excitation light illumination portion to the light detecting portion after illuminating the same with the excitation light.

A method and apparatus for detection of a particular material, such as photo-resist material, on a sample surface. A narrow beam of light is projected onto the sample surface and the fluoresced and/or reflected light intensity at a particular wavelength band is measured by a light detector. The light intensity is converted to a numerical value and transmitted electronically to a logic circuit which determines the proper disposition of the sample. The logic circuit controls a sample-handling robotic device which sequentially transfers samples to and from a stage for testing and subsequent disposition. The method is particularly useful for detecting photo-resist material on the surface of a semiconductor wafer.

The particles each of which either naturally fluoresce or are tagged to fluoresce at distinctive wavelengths are caused to flow through an analyzing volume where fluorescence is excited by an impinging light beam. A tunable optical filter repetitively and sequentially passes emitted light at each of the characteristic wavelengths as each particle travels through the analyzing volume and the light transmitted through the optical filter is received by single detector which provides output signals representative of each distinct wavelength of light emitted by the particle.

An optical imager, such as a microscope for performing multiple frequency fluorometric measurements comprising a light source, such as a laser source is disclosed. The system is used to excite a sample into the fluorescent state. Light from the excited sample is collected by a microscope. The microscope utilizes conventional confocal optics optimized to have a very narrow depth of field, thus limiting the information collected to a thin planar region. Measurements are taken over the fluorescence lifetime of the sample simultaneously from the excitation source and from the excited sample. Information is taken in a matrix and comparison of the image matrix and the standard during simultaneous measurements yields output information.

The present invention provides devices and methods for detection of particles, such as biological cells, in samples using a photosensitive waveguide. The photosensitive waveguide changes its transmissivity in a detectable manner in response to controlling radiation emitted from the particles. In preferred embodiments, the waveguide is two-dimensional and the position of the particles as well as their presence is obtained by scanning the waveguide in two non-parallel directions. The provided devices are preferably used to locate labeled cells. The present invention also includes control systems and methods for detecting and locating cells using the devices provided.

This invention concerns a fluorometer preferably combined with a thermal cycler useful in biochemical protocols such as polymerase chain reaction (PCR) and DNA melting curve analysis. The present fluorometer features a low heat-generating light source such as a light emitting diode (LED), having a one-to-one correspondence to each of a plurality of sample containers, such as capped PCR tubes in a standard titer tray. The fluorometer of the present invention further comprises an optical path between each LED and its correspondingly positioned container, and another optical path between each fluorescing sample within the positioned container and an optical signal sensing means. The instrument can be computer controlled.

Absorption wavelength spectral characteristic data of fluorescence indicators whose kinds are known and which dye the sample is acquired. The absorption wavelength spectral characteristic data of a site to be discriminated is compared with the absorption wavelength spectral characteristic data of each of the fluorescence indicators of known kinds.
