The novel tripeptides Ac-D-2Nal-D-4ClPhe-D-3Pal-OH and Boc-D-2Nal-D-4ClPhe-D-3Pal-OH are intermediates useful in the synthesis of LHRH analogs by coupling with suitable heptapeptides, in particular with the heptapeptides P1-Ser(P2)-NMeTyr(P3)-D-Lys(Nic)-Leu-Lys(iPr, P4)-Pro-D-AlaNH2 and P1-Ser(P2)-NMeTyr(P3)-D-Asn-Leu-Lys(iPr, P4)-Pro-D-AlaNH2.
Abstract:
The present invention relates to a liquid (or solution)-phase manufacturing process for preparing the decapeptide Degarelix, its protected precursor, and other useful intermediates. The invention further relates to polypeptides useful in the solution-phase manufacturing process and to the purification of Degarelix itself. Degarelix can be obtained by subjecting a Degarelix precursor according to formula (II): (P1)AA1-AA2-AA3-AA4(P4)-AA5-AA6(P6)-AA7-AA8(P8)-AA9-AA10-NH2 (II) or a salt or solvate thereof, to a treatment with a cleaving agent in an organic solvent, wherein P1 is an amino protecting groups; preferably acetyl; P4 is hydrogen or a hydroxy! protecting group, preferably a hydroxyl protecting group; P6 is hydrogen or an amino protecting groups; preferably an amino protecting groups; and P8 is an amino protecting group.

A separable coupling may be formed by a connecting stub (2) and a locking jacket (5) which, when the locking jacket is moved relative to the connecting stub, can lock a nipple (16) in the coupling if fluids under pressure are fed through the coupling, as the connecting stub is provided with a part (21, 29) therein which is movable transversely to the connecting stub, and which, when pressure is applied through the coupling, is pressed partly out of the locking jacket (5) so that movement of the locking jacket is prevented. The movable part may be formed by an O-ring (21), a four-edged ring or the like which can press a split locking ring, concentric with the O-ring, partly out of the connecting stub (2). In an embodiment, the movable part is formed by a bushing (28) with a spring-biased, movable piston (29) which presses the piston (29) out of the bushing (28) and blocks movement of the locking jacket by the action of the fluid which is fed. The bushing (28) has a central through hole in which a spring-biased piston (29) may be moved, such that when pressure is applied through the coupling, the piston protrudes through the hole of the bushing. The invention provides a coupling which ensures that the coupling parts cannot be separated unintentionally from each other, when fluids are fed through the coupling at high pressures and temperatures, which may moreover be chemically aggressive.

A method for dewatering a fibrous material web, wherein fibrous material is applied via a headbox on at least one wire and is subsequently dewatered in order to form a fibrous material web. Following the wire section, the resulting fibrous material web is dewatered by several presses, wherein the first press through which the fibrous material web is guided following the headbox is equipped with a double nip. The first press is formed by a wire roll and two couch rolls which are each pressed against the wire roll for forming a nip. Also disclosed is a corresponding device for carrying out the method.
