Abstract: Disclosed herein include methods, compositions, and kits enabling expression of multiple payload proteins from a single mRNA configured to achieve a predetermined stoichiometry of first unit payload protein(s) and second unit payload protein(s) in a cell or cell-like environment. There are provided, in some embodiments, nucleic acid compositions comprising a polynucleotide comprising a first nucleic acid unit (encoding one or more first unit payload protein(s)) and a second nucleic acid unit (encoding one or more second unit payload protein(s)). The first nucleic acid unit and the second nucleic acid unit can each comprise an engineered translation initiation site (eTIS) comprising a three-nucleotide tunable element immediately upstream of a start codon. In some embodiments, the nucleic acid composition comprises at least one modified nucleotide and/or at least one nucleotide analogue or nucleotide derivative. In some embodiments, the polynucleotide comprises a disclosed eTIS combination.

Abstract: A cranial implant that facilitates functional ultrasound imaging is disclosed. The example cranial implant replaces a section of a skull. The cranial implant in includes a support section shaped to replace a removed section of the skull. The implant includes a window of sonolucent material allowing functional ultrasound imaging through an ultrasound probe on the window. The window is shaped to allow access to a region of interest in the brain from the ultrasound probe.

Abstract: Disclosed herein include methods, compositions, and kits suitable for use in spatiotemporal regulation of probiotic cells. There are provided, in some embodiments, thermal bioswitches that allow probiotic cells to sense small changes in temperature and use them as inputs for the actuation of genetic circuits. Genetic circuits capable of inducing expression of a payload upon thermal stimulation are provided. Thermally actuated probiotic cells and methods of use for the treatment of diseases or disorders are also provided.

Abstract: Systems and methods for targeting specific cell types by selective application of ultrasonic harmonic excitation at their resonance frequency (“oncotripsy”) are presented. The systems and methods result in the permeabilization, lysis, and/or death of targeted cell types by using ultrasonic harmonic excitations that have their frequency and pulse duration specifically tuned to disrupt the nuclear membrane of the targeted cells types by inducing a destructive vibrational response therein while leaving non-targeted cell types intact. Target cells may be neoplastic.
