Leucine-rich repeat kinase-2 (LRRK2) mutations are a common cause of Parkinson's disease. Inhibitors of LRRK2 kinase that are protective in in vitro and in vivo models of LRRK2-induced neurodegeneration were identified. The presently disclosed subject matter establishes that LRRK2-induced degeneration of neurons in vivo is kinase dependent and that LRRK2 kinase inhibition provides a potential new neuroprotective paradigm for treating Parkinson's disease.

Methods for diagnosing Alzheimer's Disease by detection of fragments of nectin-1 are described. Nectin-1 is shown to be a substrate for proteases associated with the onset of Alzheimer's Disease, including α-secretase, γ-secretase and BACE1 or a BACE1-like protease. The fragments produced by the action of these and other proteases on Nectin-1 can be used to diagnosis Alzheimer's Disease or a predilection towards Alzheimer's Disease.

The invention features new helper virus-free methods for making herpes virus amplicon particles that can be used in immunotherapies, including those for treating any number of infectious diseases and cancers (including chronic lymphocytic leukemia, other cancers in which blood cells become malignant, lymphomas (e.g. Hodgkin's lymphoma or non-Hodgkin's type lymphomas). Described herein are methods of making helper virus-free HSV amplicon particles; cells that contain those particles (e.g., packaging cell lines or patient's cells, infected in vivo or ex vivo); particles produced according to those methods; and methods of treating a patient with an hf-HSV particle made according to those methods.

A triage system that determines whether a user is likely to have contracted a disease based on sensor data received from a user device (e.g., a smartphone or activity tracker). Each symptom is identified by comparing sensor data to a predetermined baseline and comparing the difference to a predetermined symptom threshold. Because direct measurement of symptoms using the sensors available to the user may not be feasible or sufficiently accurate, the triage system also uses surrogates the identify certain symptoms. For example, a fever may be identified using heart data, a cough or shortness of breath may be identified by analyzing recorded sound, fatigue may be identified by analyzing the movement of the user device, and loss of smell or taste may be identified by recording sound and using speech detection algorithms to identify phrases in the recorded sound indicative of loss of smell or taste.

An autologous vaccine to tumor cells is produced by transducing the tumor cells with a herpes simplex virus amplicon containing the gene for an immunomodulatory protein to provide transient expression of the immunomodulatory protein by the cells. The tumor cells may transduced with the herpes simplex amplicons ex vivo or in vivo. Suitable immunomodulatory proteins include cytokines, for example, interleukins, interferons, and chemokines such as RANTES; intercellular adhesion molecules, for example ICAM-1 and costimulatory factors such as B7.1. The tumor cells may also be transduced with one or more species of amplicon containing genes for two or more different immunomodulatory proteins.

Such neurodegenerative diseases include Alzheimer's disease.
