Improvements in a rope securing anchor for a boat fender that provides a versatile rope securing system to secure a boat to a dock or other location. The fender hook rope anchor can be quickly installed or removed from a boat fender and the rope can be quickly adjusted for the rope position within the fender hook rope anchor. The rope anchor has an open hook with a flexible tension finger that is easily installed and removed over the fender of a boat. A user can place the fender hook rope anchor where needed and quickly removed when not needed. There is an opening where a rope can pass and be secured using a one-way spring loaded and releasable cam. A user can quickly pass a rope through the opening to secure a line, and can pull the line out of the rope anchor when not needed.

Improvements in a line brake with strap. The brake is a one-way brake that allows a user to pull the line through the brake to shorten the length of the line and when tension in the opposite direction is placed on the line, the brake prevents the length of the line from extending through the brake. The strap is flexible and can be completely opened to freely wrap around a post, pole, line, rail, cord, capstan, handrail, cleat, post, cord or anchor, where a free end of the pole can't be easily reached over, though, under or around. The strap has a key opening that prevents the strap from easily opening when the key is not linearly aligned and/or rotated. A tail on the strap further prevents accidental dislodging of the strap from the line brake.

Assay methods may generally comprise forming homogeneous assay mixtures comprising target SAM-utilizing protein, fluorescent detection analyte, and test compound, incubating, and measuring FP or TR-FRET signal emitted in order to determine a measure of test compound-SAM-utilizing protein binding. Assay mixtures comprise a SAM-utilizing protein, and a fluorescent detection analyte that binds with the SAM-utilizing protein in the absence of test compound. Assay mixtures may further comprise a test compound. Assay mixture embodiments may generate FP or TR-FRET signal properties that are a function of the inherent binding interactions of both the test compound and the detection analyte with the SAM-utilizing protein. Fluorescent detection analytes comprise a fluorophore moiety, a covalent linker moiety, and a SAM-utilizing protein ligand moiety and could be utilized in FP or TR-FRET assays to measure test compound binding.
