Antibodies are described that are used to treat disorders associated with altered permeability of the digestive tract. Provided is methods for producing mixtures of antibodies from a single host cell clone, wherein, a nucleic acid sequence encoding a light chain and nucleic acid sequences encoding different heavy chains are expressed in a recombinant host cell. The recombinantly produced antibodies in the mixtures according to the invention suitably comprise identical light chains paired to different heavy chains capable of pairing to the light chain, thereby forming functional antigen-binding domains. Mixtures of the recombinantly produced antibodies are also provided by the invention. Such mixtures can be used in a variety of fields.

The invention provides a composition comprising polyclonal antibodies that specifically bind to with human tumor necrosis factor (hTNF) wherein the polyclonal antibodies are derived from the serum, milk or colostrum of a bovine animal that has been immunized hTNF or an immunogenic portion thereof. The antigenic specificity analysis of the anti-hTNF polyclonal antibodies of the invention is unique and has not been previously described for anti-hTNF polyclonal or monoclonal antibodies of the prior art.

The invention provides cloned transgenic ungulates (e.g., bovines) in which prion protein activity is reduced by one or more genetically engineered mutations. Desirably, these transgenic bovines are also genetically modified to express xenogenous (e.g., human) antibodies. Because of their resistance to prion-related diseases such as bovine spongiform encephalopy (also known as mad cow disease), these bovines are a safer source of human antibodies for pharmaceutical uses and a safer source of agricultural products.

The invention relates to means and methods for obtaining an antigen specific antibody from milk from non-human mammals that have not been immunized with said antigen prior to collecting said milk. Non-human mammal derived antigen specific antibody was found to cross the mucosal side of the intestinal tract of an individual. The invention provides novel means and methods for the treatment of disease through antigen specific non-human mammal antibodies. Further provided are preparations for oral administration comprising said antigen specific antibody of a ruminant.

The present invention relates to a bi-specific antibody or antibody fragment having at least one arm that is reactive against a targeted tissue and at least one other arm that is reactive against a targetable conjugate. The targetable conjugate encompasses a hapten to which antibodies have been prepared. In preferred embodiments, the hapten is histamine-succinyl-glycine (HSG). In more preferred embodiments, the at least one arm comprises the CDR sequences of the HSG-binding 679 antibody. The targetable conjugate is attached to one or more therapeutic and/or diagnostic agents. The invention provides constructs and methods for producing the bispecific antibodies or antibody fragments, as well as methods for using them and kits comprising them.
