A prism including a substrate faceted to provide a plurality of flat surfaces, wherein at least two of the plurality of surfaces, each including a filter coating, form at least two filter surfaces, wherein each filter surface selectively permits passage of a predetermined wavelength and reflects remaining wavelengths along an optical path towards another of the plurality of surfaces, optionally another filter surface, wherein an angle of incidence of each of the plurality of surfaces along the optical path is equal or nearly equal. An apparatus incorporating the prism and its use for splitting a light spectrum into a plurality of wavelengths or wavelength ranges.

A method of determining one or more beating parameters for use in cardiomyocyte beating analysis including: providing a cell analysis device including wells, each well including a sensor capable of monitoring beating of cardiomyoctes in millisecond time resolution; adding cardiomyocytes to the wells; monitoring the beating of the cardiomyocytes in millisecond time resolution to obtain a plurality of beating measurements; and calculating one or more beating parameters from the plurality of beating measurements.

The invention provides methods of investigating a mechanism of action of a compound, which includes providing a device for monitoring cell-substrate impedance; attaching the device to an impedance analyzer; adding cells to two or more wells; adding a test compound to at least one of the wells and providing at least one control well; monitoring impedance of the wells to obtain a series of impedance measurements; plotting impedance measurements to obtain impedance curves and comparing the impedance curves to determine a time frame at which the test compound has a significant effect on cell growth or behavior. Determining the time frame provides information on changes in cell status in response to the test compound, including cell attachment or adhesion status, cell growth or proliferation status, the number of viable or dead cells, cytoskeletal organization or structure, and the number of cells going through apoptosis or necrosis.

A device for detecting cells and/or molecules on an electrode surface is disclosed. The device detects cells and/or molecules through measurement of impedance changes resulting from the cells and/or molecules. A disclosed embodiment of the device includes a substrate having two opposing ends along a longitudinal axis. A plurality of electrode arrays are positioned on the substrate. Each electrode array includes at least two electrodes, and each electrode is separated from at least one adjacent electrode in the electrode array by an expanse of non-conductive material. The electrode has a width at its widest point of more than about 1.5 and less than about 10 times the width of the expanse of non-conductive material. The device also includes electrically conductive traces extending substantially longitudinally to one of the two opposing ends of the substrate without intersecting another trace. Each trace is in electrical communication with at least one of the electrode arrays.

A method of identifying a therapeutic compound for treating cancer in a human subject, the method including: providing a device that measures cell-substrate impedance; culturing cancer cells in the at least two wells, wherein the cancer cells are obtained from a human subject and have a receptor tyrosine kinase (RTK) pathway; adding to a first well a proposed therapeutic compound that affects an RTK pathway and an RTK stimulating factor for the RTK pathway to form a test well, and adding to another well the RTK stimulating factor to form a control well; continuously monitoring cell-substrate impedance of the at least two wells; and determining a difference in impedance or optionally in cell index between the test well and control well; and if significantly different, concluding the proposed therapeutic compound is therapeutically active in the RTK pathway within the cancer cells of the human subject.

An optical engine its use in a bench top flow cytometer, the optical engine having a set of lasers, each focused horizontally along an x-axis to a same horizontal position and vertically along a y-axis to a different vertical position along a same excitation plane of a flow cell, a set of optics that separate fluorescence of a same wavelength range into different locations in a focal plane of collection optics according to the different lasers by which the fluorescent light is excited; and a detector that selectively detects light from the different locations thereby distinguishing between fluorescence emitted within the same wavelength range as excited by the different lasers.
