Abstract: A method for packing a media bed in a column (3). According to the invention the method comprises the steps of: providing data to a control unit (15) connected to the column (3), said data comprising at least a measured slurry concentration, a target bed height, a target packing factor or compression factor and minimum and maximum acceptable values for at least one of target bed height and target packing or compression factor; forcing a movable adapter (9) along a longitudinal axis of the column (3) in order to pack the media; detecting when the media bed is consolidated; the control unit (15) processing the provided data and the information about the consolidated bed height in order to present to the user acceptable bed heights, if there are any, giving acceptable packing or compression factors.

Abstract: A method of operating a chromatography column is described for use in methods of manufacture for producing anti-IL-12/IL-23p40 antibodies, e.g., the anti-IL-12/IL-23p40 antibody STELARA® (ustekinumab). This method involves collecting column outlet signal and accumulated flow parameters at two or more intervals of at least one mobile phase transition front during operation of the chromatography column comprising column packing. A model gamma cumulative distribution curve is calculated based on the collected column outlet signal and accumulated flow parameters for the at least one mobile phase transition front. A height equivalent theoretical plate (HETP) value is calculated for the at least one mobile phase transition front using parameters of the model gamma cumulative distribution curve and the quality of the chromatography column packing is assessed based on the calculated HETP value.

Abstract: The invention provides methods for preparing a polymer-grafted and functionalized nonwoven membrane adapted for use in separation processes. The invention further provides so-formed membranes as well as improved separation methods utilizing the membranes. The polymer-grafted and functionalized nonwoven membranes are particularly formed utilizing thermal grafting. In particular, an acrylate or methacrylate polymer can be grafted onto a nonwoven web comprising a plurality of polymeric fibers to form a plurality of polymer segments covalently attached to the polymeric fibers. Thermal grafting particularly can comprise using a thermal initiator and exposing the nonwoven web to heat to initiate polymerization of the acrylate or methacrylate monomer. The grafted polymeric fibers can be functionalized to attach at least one functional group adapted for binding to a target molecule to the polymer segments of the grafted polymeric fibers.

Abstract: The present invention aims at reducing the time required for a series of analyses in the sequential performance of gradient analyses under a variety of conditions. To this end, in a control apparatus for controlling the operation of a liquid chromatograph having a gradient analysis function in which a mobile phase composed of a plurality of mixed solvents is used and a chromatograph analysis is performed while the mixture ratio of the solvents is temporally changed, the liquid chromatograph is controlled so as to continuously change the mixture ratio of the solvents from an initial mixture ratio to a final mixture ratio when performing a sample analysis; and as to perform, before the sample analysis, a preparatory liquid supply in which the mixture ratio of the solvents is continuously changed from the initial mixture ratio to the final mixture ratio at a rate higher than that in the sample analysis.
