Disclosed are a non-human mammalian cell, a recombinant genome thereof, and a method for producing same. The variable region genes of the endogenous immunoglobulin in the genome are partially or entirely replaced with variable region genes of human immunoglobulin, a part or all of the pseudogenes and/or open reading frames of the variable region genes of human immunoglobulin, including coding and non-coding sequences for human heavy chain functional VH, DH, JH, or coding and non-coding sequences for human light chain functional VL, JL, are knocked out. A non-human mammalian cell of the present invention that contains human immunoglobulin domains can be used to produce a transgenic animal that is capable of producing antibodies with fully human variable domain(s), whereby fully human antibodies featuring a higher affinity can be screened efficiently at a lower cost within a shorter period.

Herein is reported a method for the humanization of non-human antibodies using a structure-based scoring matrix. With the scoring matrix it is possible to determine (the requirement for and) the suitability of specific (back)mutations of amino acid residues at defined positions of a selected human germline sequence. The scoring matrix takes into account the topology, the three-dimensional structure and the interactions of the respective residue and change. Thereby the influence on antigen binding of a specific amino acid residue change can be determined.

Abstract:
The disclosure provides a method for generation of humanized full length antibodies in mammalian cells. A library of humanized variants is provided with high, validated human framework diversity without requiring back-mutations to retain original affinity. Synthetic CDR encoding fragment libraries derived from a template antibody are ligated to human framework region encoding fragments from a human framework pool limited only to germline sequences from a functionally expressed antibodies. The vector comprises a nucleic acid sequence encoding HC framework region 4. No CDR grafting or phage display is required.

The present invention discloses antibodies which bind human and Macaca fascicularis CEACAM5 proteins, as well as isolated nucleic acids, vectors and host cells comprising a sequence encoding said antibodies. The invention also discloses immunoconjugates comprising said antibodies conjugated or linked to a growth-inhibitory agent, and to pharmaceutical compositions comprising antibodies, or immunoconjugates of the invention. The antibodies or immunoconjugates of the invention are used for the treatment of cancer or for diagnostic purposes.

The present invention relates to immunoglobulins that bind IgE and FcγRIIb with high affinity, said compositions being capable of inhibiting cells that express membrane-anchored IgE. Such compositions are useful for treating IgE-mediated disorders, including allergies and asthma.
