Abstract: The present disclosure provides variant major histocompatibility complex class II (MHCII) beta chains, as well as heterodimers and multimers including MHCII alpha chains and the variant MHCII beta chains. Also provided by the present disclosure are nucleic acids, expression cassettes, and expression vectors encoding the MHCII beta chains. The heterodimers and multimers of the present disclosure have a higher affinity for CD4 co-receptors than comparable reagents including wild type MHCII beta chains, and therefore are advantageous for use in methods of phenotyping or activating CD4+ T cells.

Abstract: Methods of quantifying anti-HPA-1a antibodies in a sample, comprising (a) contacting the sample with a capture reagent that binds to the anti-HPA-1a antibodies to form a detection complex, wherein the capture reagent comprises a modified integrin psi and (b) measuring an amount of the detection complex, wherein the amount of the detection complex indicates the quantity of the anti-HPA-1a antibodies in the sample. The methods may also comprise contacting the sample with a detecting reagent that binds to the anti-HPA-1a antibodies and becomes part of the detection complex. The capture reagent and/or the detecting reagent may comprise a detection label, in which measuring the amount of the detection complex comprises measuring the amount of the detectable label.

Abstract: The present invention describes kits and methods for rapid detection of nucleic acid sequences in real-time and in any setting. Unique features of this kit include isothermal nucleic acid extraction in Unit 1, and amplification, labeling, and signal detection in Unit 2 (FIG. 1). These units can be operated separately with manual transfer of product from one to the other or can be put together in a fashion where user places sample into inlet of Unit 1 and observes results in detection chamber of Unit 2. Several versions of this kit can be developed using different sequence-specific primers and probes to detect any nucleic acid sequence (DNA or RNA) in a multiplex fashion. In a preferred embodiment, visual detection of amplified signal is done using Lateral Flow Strips, eliminating the need for any additional complex equipment. Such a kit can be easily developed and manufactured to assist in the mitigation response to any pandemic, especially for COVID-19. The COVID-19 kit is called an At-Home COVID test (AH-COV).

Abstract: A non-invasive system and method for disease detection and real-time monitoring of organ health wherein the changes in the electrical properties of urine samples are studied over time in the presence or absence of interacting chemicals for qualitative and quantitative estimation of urinary analytes. The system has two components—a test platform and a reader. The test platform is integrated with electrodes and chemicals, which interact with urine to detect the biomarker of the disease and quantify the analyte. The changes in the electrical properties serve as an electrical signature for a particular analyte. This electrical signal from the test platform is relayed to an electronic reader that receives, processes, and analyzes the data for single test detection or continuous monitoring. The reader stores the information that can then be coupled to a readout system or transmitted by wired or wireless mechanisms to any electronic platform in real-time.
